Siglec-15与DUSP1在肝细胞癌中的表达关系及其临床意义

The expression relationship and clinical significance of Siglec-15 and DUSP1 in hepatocellular carcinoma

目的分析Siglec-15和DUSP1在肝细胞癌(hepatocellutar carcinoma,HCC)中的表达及两者之间的关系以及其与临床病理因素、预后的关系。方法收集2021年6月至2022年6月于郑州大学附属洛阳中心医院行肝癌根治切除术的56例HCC患者癌组织及正常组织石蜡切片标本。免疫组化检测Siglec-15和DUSP1的表达,构建对照组(Control)、空载体组(NC)、高表达Siglec-15(Siglec-15)、低表达Siglec-15(sh-Siglec-15),利用qRT-PCR、Western blotting检测Siglec-15和DUSP1的表达。结果HCC组织中Siglec-15蛋白高于正常组织(P<0.001),DUSP1蛋白低于正常组织(P<0.001),且Siglec-15蛋白表达与DUSP1蛋白表达呈负相关(r=-0.31,P<0.05),构建Siglec-15低表达细胞株,发现sh-Siglec-15组HCC细胞中的DUSP1的表达升高(P<0.05),构建Siglec-15高表达细胞株,发现Siglec-15组HCC细胞中的DUSP1的表达降低(P<0.05);Siglec-15的表达与患者的TNM分期相关,DUSP1的表达与患者的TNM分期及AFP值相关(P<0.05);Siglec-15和DUSP1表达与患者1年无进展生存期相关。结论DUSP1在HCC组织中低表达,与Siglec-15的表达呈负相关;Siglec-15可能通过DUSP1控制下游通路。

Objective To analyze the expression and their relationship of Siglec-15 and DUSP1 in hepatocellular carcinoma(HCC),and explore their clinical pathological factors and prognostic value.Methods A total of 56 HCC patients who underwent radical resection of liver cancer at Luoyang Central Hospital Affiliated to Zhengzhou University from Jun.2021 to Jun.2022 were collected.Cancer tissue and normal tissue paraffin sections were collected and subjected to immunohistochemistry to detect the expression of Siglec-15 and DUSP1.Control group(Control),empty body group(NC),high expression Siglec-15(Siglec-15),low expression Siglec-15(sh-Siglec-15)were constructed,and the expressions of DUSP1 and Siglec-15 were detected by qRT-PCR and Western blotting.Results The Siglec-15 protein in HCC tissue was significantly higher than that in normal tissue(P<0.001).The expression of DUSP1 protein in HCC tissue was significantly lower than that in normal tissue(P<0.001)and there was a negativel relation between the expression of siglec-15 protein and DUSP1 protein(r=-0.31,P<0.05).A low expression cell line of Siglec-15 was constructed,and it was found that the expression of DUSP1 in sh-Siglec-15 group HCC cells was increased(P<0.05).A high expression cell line of Siglec-15 was constructed,and it showed that the expression of DUSP1 in Siglec-15 group HCC cells was reduced(P<0.05).The expression of Siglec-15 was correlated with the TNM stage of patients,while the expression of DUSP1 was correlated with the TNM stage and AFP value of patients(P<0.05).The expression of Siglec-15 and DUSP1 was associated with one-year progression-free survival of patients.Conclusion DUSP1 is low expressed in HCC tissue and negatively correlated with the expression of Siglec-15.Siglec-15 may control downstream pathways through DUSP1.
