摘要
补体系统和凝血系统有共同的起源,可以在不同的细胞中相互作用。它们的相互作用在免疫、凝血和各种疾病中都很重要,包括癌症和2019年爆发的冠状病毒病(新冠肺炎)。通过共享受体的表达和细胞因子的分泌,内皮细胞和血小板等极大地增强了两个系统之间的沟通。尽管已有许多研究描述了这两个系统之间细胞介导的相互作用,但仍需要更深入的理解。本文旨在总结内皮细胞、血小板、多形核白细胞(polymorphonuclear leukocytes,PMNs)和单核吞噬细胞系统(mononuclear phagocyte system,MPS)这两个系统之间分子通讯的最新知识,关注两个系统如何通过在其表面招募共享调节因子来防止过度激活。通过提供这两个系统之间细胞特异性串扰的最新概述,本文可能有助于探索疾病相关的分子事件、药物开发和靶向治疗。
The complement and coagulation systems share a common origin and can interact in various cells.Their interactions are important in immunity,blood coagulation and various diseases,including cancer and coronavirus disease of 2019(COVID-19).Through the expression of shared receptors and secretion of cytokines,cells such as endothelium and platelets greatly enhance the communication between the two systems.Although the cell-mediated crosstalk between the two systems has been described,a deeper understanding is still needed.This review aims to summarize the current knowledge on the molecular communication between the two systems in endothelium,platelets,polymorphonuclear leukocytes(PMNs),and the mononuclear phagocyte system(MPS).It focuses on how the two systems prevent excessive activation by recruiting shared regulators on their surface.By providing an updated overview of the cell-specific crosstalk between the two systems,this work may contribute to the exploration of disease-related molecular events,drug development,and targeted therapies.
