土鳖虫指纹图谱分析

摘要 土鳖虫是一种动物类中药,入药部位为鳖蠊科昆虫地鳖Eupolyphaga sinensis Walker或冀地鳖Steleophaga plancyi Boleny的雌虫干燥体,而雄虫不能入药,为保证用药的安全性和有效性,需准确有效区分雌雄土鳖虫。本研究采用高效液相色谱法,色谱柱采用Zafex Supperfex AQ-C_(18)柱(4.6 mm×250.0 mm,5μm),以甲醇为流动相A,以0.1%冰醋酸溶液为流动相B,梯度洗脱60 min,柱温30℃,检测波长248 nm,进样量10μL。结果显示,28批土鳖虫(雌虫)均呈现与土鳖虫对照药材相对应的8个特征峰色谱峰,以对照品指认了其中4个化学成分,分别为尿嘧啶、次黄嘌呤、黄嘌呤、鸟苷,且与土鳖虫雄虫的指纹图谱不匹配。该方法可以有效区分雌雄土鳖虫,有效避免掺伪情况发生,为土鳖虫质量控制提供了参考方法。 Tubiechongis a dried female of Eupolyphaga sinensis Walker or Steleophaga plancyi Boleny,while the male worm can not be used as medicine.In order to improve the safety and effectiveness of drugs,it should to accurately and effectively distinguish the male and female of Tubiechong.High performance liquid chromatography (HPLC) was performed on Zafex SupperfexAQ-C_(18)(4.6 mm×250.0 mm,5μm) column.Mobile phase A was methanol,and mobile phase B was 0.1%glacial acetic acid solution in gradient elution of 60 min.The column temperature was 30℃.The detection wavelength was 248 nm and sample volume was 10μL.The results showed that the chromatographic peaks of8 characteristic peaks corresponding to the control materials of 28 batches of Tubiechong (female) were found4 chemical components were identified by the control materials,namely uracil,hypoxanthine,xanthine and guanosine And it did not match the fingerprint of the male.This method can effectively distinguish male and female of Tubiechong and can effectively avoid adulteration,which can provide reference method for quality control of Tubiechong.
