AURKA在肺腺癌中的表达及其临床意义的研究

Expression and clinicopathologic significance of AURKA in lung adenocarcinoma

目的 研究AURKA在肺腺癌中的表达及其临床意义。方法 回顾性收集2020年6月-2022年6月期间于湖北民族大学附属荆门市第一人民医院胸外科接受手术治疗的68例肺腺癌患者的手术标本,包括癌组织和癌旁组织。采用生物信息学技术分析,实时荧光定量PCR(qPCR)、Western Blot和免疫组化染色验证相结合的方法,分别检测并比较肺腺癌与癌旁组织、正常支气管粘膜上皮细胞与肺腺癌细胞株AURKA mRNA及其蛋白的表达丰度;分析肺腺癌组织中AURKA表达水平与患者临床特征的关系;并探讨AURKA在肺腺癌发生发展中的作用及其潜在的机制。结果 肺腺癌组织中AURKA的表达量显著高于其癌旁组织,差异有统计学意义(P<0.01);且AURKA表达量与肺腺癌TNM分期、病理分化程度以及淋巴结转移相关,差异有统计学意义(P<0.05);AURKA通过影响AURKA/PLK1信号轴,调控肺腺癌细胞增殖、侵袭,促进肺腺癌的发生与发展。结论 AURKA在肺腺癌的发生和发展过程中发挥着重要作用,可作为肺腺癌患者预后及复发的预测指标,并可能成为肺腺癌治疗的潜在靶点。

Objective To study the expression of AURKA in lung adenocarcinoma and its clinical significance.Methods From June 2020 to June 2022,68 patients with lung adenocarcinoma underwent surgical treatment in the Department of Thoracic Surgery of the First People's Hospital of Jingmen Affiliated to Hubei University for Nationalities,and the surgical specimens of cancerous tissue and the paracancerous tissue were collected retrospectively.The expression abundance of AURKA mRNA and protein in lung adenocarcinoma and the paracancerous tissues,normal bronchial epithelial cells and lung adenocarcinoma cell lines were detected and compared by bioinformatics analysis,qPCR,Western blot and immunohistochemical staining.The relationship between the expression level of AURKA in lung adenocarcinoma and the clinical characteristics of patients was analyzed,and the role of AURKA in the development of lung adenocarcinoma and its potential mechanism were explored.Results The expression level of AURKA in lung adenocarcinoma tissue was significantly higher than that in its adjacent tissues(P<0.01).The expression level of AURKA was significantly correlated with tumor TNM stage,pathological differentiation and lymph node metastasis of lung adenocarcinoma(P<0.01).AURKA regulated the proliferation and invasion of lung adenocarcinoma and promoted the occurrence and development of lung adenocarcinoma through modulating the AURKA/PLK1 axis.Conclusion AURKA plays an important role in the pathogenesis and evolution of a subset of lung adenocarcinoma,which can be used as a predictor of prognosis and recurrence of lung adenocarcinoma,and might be a potential target for the treatment of lung adenocarcinoma.
