“健脾益气和胃法”电针对糖尿病胃轻瘫大鼠胃平滑肌细胞凋亡的影响研究

The Electroacupuncture Effect of“Invigorating Spleen and Invigorating Qi and Stomach”on Apoptosis of Gastric Smooth Muscle Cells in Diabetic Gastroparesis Rats

目的:研究“健脾益气和胃法”电针对糖尿病胃轻瘫(DGP)大鼠胃肠运动恢复及其胃平滑肌细胞(SMCs)凋亡的影响。方法:采用腹腔注射链脲佐菌素(STZ)后普通饲料常规喂养8周的方法建立DGP大鼠模型,应用“健脾益气和胃法”电针治疗,治疗4周后,检测各组大鼠体质量、空腹血糖(FBG)、胃肠运动情况;应用HE与Tunel染色,观察各组大鼠胃组织形态及其SMCs凋亡情况;应用qRT-PCR法检测各组大鼠胃组织中Bcl-2、Bax、Caspase-3 mRNA的表达情况。结果:与模型组比较,“健脾益气和胃法”电针可促进DGP大鼠精神状态、毛色、饮食、饮水、大便、小便等一般情况的恢复,降低DGP大鼠FBG水平,增强DGP大鼠胃排空率,并可促进DGP大鼠胃组织的形态修复,抑制SMCs凋亡,降低Bax、Caspase-3 mRNA的表达,增加Bcl-2 mRNA的表达。结论:“健脾益气和胃法”电针可促进DGP大鼠胃肠运动的恢复,并可抑制SMCs凋亡,从而有效治疗糖尿病胃轻瘫。

Objective:To study the restoration of gastrointestinal movement and its effects on apoptosis of gastric smooth muscle cells(SMCs)in rats with the“invigorating spleen and Qi and the stomach method”by Diabetic Gastroparesis(DGP).Methods:DGP rat models were established by intraperitoneal injection of Streptozocin(STZ)followed by routine feeding of ordinary feed for 8 weeks.Electroacupuncture was used to strengthen spleen and Qi and stomach.After 4 weeks of treatment,body weight and Fasting blood glucose of the rats were measured.HE and Tunel staining were used to observe the gastric morphology and SMCs apoptosis of rats in each group.mRNA expressions of Bcl-2,Bax and Caspase-3 in gastric tissues were detected by qRT-PCR.Results:Compared with model group,“invigorating spleen and invigorating Qi and stomach method”electroacupuncture can promote the recovery of mental state,hair color,diet,drinking water,stool,urination and other general conditions of DGP rats,reduce FBG level of DGP rats,enhance gastric emptying rate of DGP rats,promote the morphological repair of gastric tissue,inhibit SMCs apoptosis of DGP rats.The expression of Bax and Caspase-3 mRNA was decreased,and the expression of Bcl-2 mRNA was increased.Conclusion:“Invigorating spleen and invigorating Qi and stomach”electroacupuncture can promote the recovery of gastrointestinal motility and inhibit SMCs apoptosis in DGP rats,so as to be effective in the treatment of DGP.
