针刺通过RA/TGF-β1/MMPs信息通路干预大鼠近视机制研究

摘要 目的:探讨针刺通过RA/TGF-β1/MMPs信息通路干预大鼠近视的机制。方法:选取60只健康1周龄大鼠作为研究对象,随机分为针刺组、模型组及对照组,每组各20只。其中对照组不予干预,针刺组和模型组通过半透明乳胶头套诱导右眼制备形觉剥夺性近视(FDM)模型,针刺组在建模完成当天,在每日当天对大鼠进行针刺干预治疗,时间2周。比较各组大鼠实验前后右眼屈光度及眼轴长度变化,观察各组大鼠巩膜形态病理学变化,HPLC法测定视网膜RA含量,在大鼠巩膜中,通过Western Blot技术检测MMP-2、TIMP-2和TGF-β1蛋白的表达水平。结果:与正常对照组相比,模型组大鼠巩膜变薄,原蛋白纤维呈无序、稀疏状分布,纤维间隙增大。针刺组巩膜增厚,胶原纤维分离减轻,排列趋向有序。与对照组相比,模型组和针刺组实验前后的屈光度明显降低,眼轴长度明显升高;而与模型组相比,针刺组大鼠屈光度明显升高,眼轴长度明显降低,差异具有统计学意义(P<0.05)。与对照组相比,模型组和针刺组大鼠巩膜中MMP-2、TIMP-2、TGF-β1蛋白表达及视网膜RA含量均明显升高,而与模型组相比,针刺组大鼠上述指标显著降低,差异具有统计学意义(P<0.05)。结论:针刺可通过对FDM大鼠视网膜组织中RA进行调节,从而间接地影响巩膜组织中TIMP-2、MMP-2及TGF-β1的表达,延缓近视的发生发展,具有一定的临床参考价值。 Objective:Exploring Acupuncture Through RA/TGF-β1/MMPs information pathway intervention in myopia in rats.Methods:60 healthy 1-week-old rats were selected as the study subjects and randomly divided into acupuncture group,model group,and control group,with 20 rats in each group.The control group was not intervened,while the acupuncture group and model group were induced to develop a form deprivation myopia(FDM)model in the right eye using a semi transparent latex headset.On the day of modeling completion,the acupuncture group received acupuncture intervention treatment on rats daily for 2 weeks.Compare the changes in right eye refractive index and axial length of each group of rats before and after the experiment,observe the morphological and pathological changes of the sclera in each group of rats,determine the content of retinal RA by HPLC,and detect MMP-2,TIMP-2,and TGF-β1 in the rat sclera using Western Blot technology-β1 protein.Results:Compared with the normal control group,the sclera of the model group rats became thinner,with disordered and sparse distribution of original protein fibers and increased fiber gaps.The acupuncture group showed thickening of the sclera,reduced separation of collagen fibers,and an orderly arrangement.Compared with the control group,the refractive index of the model group and the acupuncture group decreased significantly before and after the experiment,and the axial length of the eye increased significantly;Compared with the model group,the acupuncture group showed a significant increase in refractive index and a significant decrease in axial length(P<0.05).Compared with the control group,MMP-2,TIMP-2,and TGF-β1 in the sclera of rats in the model group and acupuncture group-βThe protein expression and retinal RA content were significantly increased in the acupuncture group,while compared with the model group,the above indicators in the acupuncture group rats were significantly reduced(P<0.05).Conclusion:Acupuncture can indirectly affect TIMP-2,MMP-2,and TGF-β1 in the scleral tissue of FDM rats by regulating RA in the retinal tissue-βThe expression of myopia can delay the occurrence and development of myopia,and has certain clinical reference value.
