鱼腥草芩蓝合剂HPLC指纹图谱及8种成分含量测定

摘要 目的:建立鱼腥草芩蓝合剂的HPLC指纹图谱,并同时测定其中黄芩苷、汉黄芩苷、汉黄芩素、连翘苷、连翘酯苷A、绿原酸、芦丁和槲皮苷的含量。方法:采用SHIMSEM Ankylo C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相为甲醇-0.1%磷酸,梯度洗脱;检测波长210 nm;体积流量1.0 mL/min;柱温40℃;进样量10μL。以黄芩苷作为参照峰,建立17批鱼腥草芩蓝合剂的指纹图谱,综合应用相似度评价、聚类分析和主成分分析评价鱼腥草芩蓝合剂批间质量差异;并同时测定其中8种成分的含量。结果:17批样品中有27个共有峰,相似度均>0.990。聚类分析可将样品分为5类,筛选出3个主成分,累计贡献率75.804%。8种成分在各自范围内线性关系良好(r≥0.9995),平均加样回收率为96.17%~103.53%,RSD为0.50%~1.23%。结论:该试验方法稳定、可靠,可对鱼腥草芩蓝合剂进行质量控制。 Objective:To establish the HPLC fingerprint of Yuxingcao Qinlan Mixture,and to simultaneously determine the contents of baicalin,wogonoside,wogonin,phillyrin,forsythoside A,chlorogenic acid,rutin and quercitrin.Methods:The SHIMSEM Ankylo C_(18) column(4.6mm × 250mm,5μm) was used,and the mobile phase was methanol-0.1% phosphoric acid in a gradient elution.The detection wavelength was 210nm,the flow rate was1.0 mL/min,the column temperature was 40℃,and the injection volume was 10μL.Using baicalin as reference,HPLC fingerprints of 17 batches of Yuxingcao Qinlan Mixture were established.Similarity analysis,hierarchical cluster analysis and principal component analysis were used to evaluate the quality difference between batches of Yuxingcao Qinlan Mixture.The contents of 8 constituents were simultaneously determined.Results:There were 27 common peaks in the 17 batches of Yuxingcao Qinlan Mixture,and the similarity were all more than 0.990.The samples were classified into 5 types according to similarity analysis.3 principal component factors were selected,and the cumulative contribution rate was 75.804%.8 constituents showed good linear relationships within their own ranges(r≥0.999 5),and the average recovery were 96.17%~103.53%,RSD were0.50%~1.23%.Conclusion:The method is stable and reliable,which can be used for quality control of Yuxingcao Qinlan Mixture.
