摘要
目的:通过检测水通道蛋白1(Aquaporin 1,AQP1)、水通道蛋白5(Aquaporin 5,AQP5)在变应性鼻炎(AR)大鼠血清及鼻黏膜中表达的变化,观察苓桂术甘汤对AR大鼠鼻腔黏膜高分泌的影响。方法:通过建立AR大鼠模型,并将大鼠随机分为模型组、治疗组(氯雷他定组及苓桂术甘汤组),另设空白组,每组10只。治疗组分别给予相应药物灌胃,空白组和模型组给予生理盐水灌胃。灌胃结束后,记录大鼠行为学积分;采用ELISA检测血清AQP1、AQP5含量及血清免疫球蛋白E(Immunoglobulin E,IgE)、卵清蛋白特异性IgE(Ovalbumin specific immunoglobulin E,OVA-sIgE)水平;采用qRT-PCR检测鼻黏膜AQP1、AQP5 mRNA表达。结果:与空白组比较,模型组大鼠行为学积分升高(P<0.05),血清AQP1、AQP5含量及IgE、OVA-sIgE水平升高(P<0.05),鼻黏膜AQP1、AQP5 mRNA表达上调(P<0.05)。治疗后,与模型组比较,治疗组大鼠行为学积分降低(P<0.05),血清AQP1、AQP5含量及IgE、OVA-sIgE水平降低(P<0.05),鼻黏膜AQP1、AQP5 mRNA表达下调(P<0.05)。结论:苓桂术甘汤可通过下调AQP1、AQP5的表达,改善鼻腔黏膜水液代谢,恢复鼻腔黏膜组织的生理功能,从而缓解AR症状。
Objective:Observe the effect of Linggui Zhugan Decoction on nasal hypersecretion in allergic rhinitis(AR) rats,through the levels of Aquaporin 1(AQP1) and Aquaporin 5(AQP5).Methods:AR model was established by ovalbumin(OVA) sensitization in SD rats.AR rats successfully modeled were randomly divided into model group,loratadine group,and Linggui Zhugan Decoction group.Another blank group was set.There were 10 rats in each group.After modeling,corresponding drugs were given.The rats in blank group and model group were given normal saline instead.Observe the behavior change of rats in each group;ELISA was used to detect the levels of immunoglobulin E(IgE),ovalbumin specific immunoglobulin E(OVA-sIgE) and AQP1,AQP5 in serum;qRT-PCR was used to detect the expression of AQP1 and AQP5 mRNA in nasal mucosa.Results:Compared with blank group,the symptoms score,levels of IgE,OVA-sIgE and AQP1,AQP5 in serum,and the expression of AQP1,AQP5 mRNA in nasal mucosa were significantly increased in model group(P<0.05).Compared with model group,the symptoms score,levels of IgE,OVA-sIgE and AQP1,AQP5 in serum,and the expression of AQP1,AQP5 mRNA in nasal mucosa were significantly decreased in Linggui Zhugan Decoction group(P<0.05).Conclusion:Linggui Zhugan Decoction could improve water metabolism of nasal mucosa by down-regulating the expression of AQP1 and AQP5,thus alleviating the symptoms on AR.
