黄蜀葵花药材、饮片、标准汤剂与配方颗粒的特征图谱相关性研究及量值传递分析

摘要 目的:建立黄蜀葵花药材、饮片、标准汤剂与配方颗粒的高效液相色谱(UPLC)特征图谱,并同时测定黄酮类成分的含量,考察四者之间化学成分的传递与差异。方法:采用Shim-packGISTC_(18)-AQ色谱柱(2.1 mm×150 mm,1.9μm),以乙腈(A)和0.1%磷酸溶液(B)为流动相梯度洗脱,检测波长为360 nm,流速为0.30 mL·min^(-1),柱温为30℃。以含量、特征图谱共有峰传递数及相似度为主要评价指标,分析其量值传递规律。结果:18批黄蜀葵花药材、饮片、标准汤剂及3批配方颗粒特征图谱均呈现8个共有特征峰,与各自对照特征图谱相似度均>0.90,辨别出杨梅素-3-O-β-D-葡萄糖苷(峰2)、金丝桃苷(峰3)、异槲皮苷(峰4)、棉皮素-8-O-β-D-葡萄糖醛酸苷(峰5)、杨梅素(峰6)、槲皮素-3′-O-葡萄糖苷(峰7)、槲皮素(峰8)7个成分,并同时测定其含量,未有离散数据出现。结论:黄蜀葵花药材、饮片、标准汤剂与配方颗粒的主要化学成分基本相同,表明特征图谱具有较高的相关性。7个成分在测定范围内有良好的线性关系,所建立的整体质量评价方法系统能够客观、全面地反映黄蜀葵花4种形态多个成分的综合情况,为黄蜀葵花配方颗粒后续质量标准的控制提供一定的参考依据。 Objective:To establish the characteristics of Radix Althaeae Sunflower,decoction pieces,standard decoction and formula granules by high performance liquid chromatography(UPLC),and to determine the content of flavonoids simultaneously,and to investigate the transfer and difference of chemical components among the four.Methods:The chromatography was performed on A Shim-packGISTC_(18)-AQ Column(2.1mm×150mm,1.9μm) with acetonitrile(A) and 0.1% phosphoric acid solution(B) as mobile phase gradient elution.The detection wavelength was 360nm,the flow rate was 0.30mL· min^(-1),and the column temperature was 30℃.Taking content,common peak transfer number and similarity of characteristic map as the main evaluation indexes,the law of magnitude transfer was analyzed.Results:There were 8 common characteristic peaks in the characteristic maps of 18 batches of Radix Althaeae Sunflower.decoction pieces,standard decoction and 3 batches of formula granules,and the similarity between them and the control characteristic maps was greater than 0.90.Myricetin-3-O-β-D-glucoside(peak 2),hypericin(peak 3),isoquercetin(peak 4),gossypitin-8-O-β-D-glucuronide(peak 5),myricetin(peak 6),quercetin-3′-O-glucoside(peak 7),quercetin(peak 8) were identified and their contents were determined simultaneously.No discrete data appears.Conclusion:The main chemical components of the herbs,decoction pieces,standard decoction and formula granules are basically the same,indicating that the characteristic map has a high correlation.The 7 components have good linear relationship in the determination range.The established overall quality evaluation system can objectively and comprehensively reflect the comprehensive appearance of the 4 forms and multiple components of the Radix Althaeae Sunflower,and provide a certain data basis for the subsequent quality control of the Radix Althaeae Sunflower formula granules.
