摘要
[目的]比较8个品牌粉末MS培养基的使用效果,为植物组织培养选购培养基提供参考。[方法]以紫锥菊的叶片、叶柄、根以及金线莲的茎段为外植体,以标准MS和不含有易制爆化合物的国家授权发明专利培养基为对照,比较8个品牌的粉末MS培养基在紫锥菊和金线莲愈伤组织诱导、不定芽再生以及生根能力的作用。[结果]粉末MS培养基中的3个品牌,代号M_(5)、M_(6)及M_(8)对紫锥菊外植体的愈伤组织诱导效果较好,且与2个对照(CK_(1)、CK_(2))相比无显著差异,而其他5个品牌的培养效果均显著差于对照。对紫锥菊不定芽诱导试验的结果显示,品牌代号M_(1)、M_(2)、M_(3)、M_(4)和M_(7)的5种培养基无论以叶片还是以叶柄为外植体均未成功诱导获得不定芽,而M_(5)培养基则不仅成功诱导获得不定芽,且获得的不定芽数均多于对照。在金线莲的组培比较试验中,10种培养基均成功诱导获得不定芽,且培养基M_(4)的不定芽诱导率高于对照组,达83.5%。诱导紫锥菊不定芽生根的比较结果显示,仅M_(5)和M_(8)这2个品牌能够和2个对照一样成功诱导紫锥菊不定芽产生不定根,但M_(5)诱导获得的不定根数和不定根长均低于2个对照。在诱导金线莲侧芽生根的试验中,8个品牌的培养基均能成功诱导产生不定根,且M_(2)诱导获得的不定根数和不定根长均显著高于对照。[结论]8个品牌的粉末MS培养基在紫锥菊和金线莲组培中的使用效果存在显著差异,且除M_(5)、M_(6)和M_(8)外,其他品牌的粉末MS培养基均不适用于紫锥菊的组织培养。但对金线莲而言,除M_(4),其他7个品牌的粉末MS培养基均适用。
[Objective]To compare the effectiveness of 8 brands of powder MS medium,so as to provide a reference basis for selecting medium for plants tissue culture.[Method]Using the leaves,petioles,roots of E.purpurea and stem segments of A.roxburghii as explants,the standard MS and the national authorized invention patent medium without explosive compounds were used as controls,the effectiveness of 8 brands of powder MS medium on callus induction,adventitious bud regeneration,and rooting of E.purpurea and A.roxburghii was compared.[Result]The three brands of powder MS medium codes M_(5),M_(6) and M_(8) were more effective in callus induction of E.purpurea,and no significant difference compared with the two controls(CK 1 and CK 2),whereas the other five brands were significantly worse than the control.The results of the experiment on inducing adventitious bud of E.purpurea showed that five brands medium codes M_(1),M_(2),M_(3),M_(4) and M_(7) failed to induce adventitious buds either by using leaves or petioles as explants in E.purpurea,while the M_(5) medium not only successfully induced but also obtained more adventitious buds than the control.On the other hand,the comparative experiment of A.roxburghii tissue culture showed that all 10 mediums could successfully induce to obtain adventitious buds,and the induction rate of adventitious buds in M_(4) was higher than the control,reaching 83.5%.The results of inducing adventitious root in E.purpurea showed that only the medium codes M_(5) and M_(8) could successfully induce adventitious roots from adventitious buds as well as the two controls,but the number and length of adventitious roots induced by M_(5) medium were lower than the two controls.In the experiment of inducing lateral bud rooting of A.roxburghii,all 8 brands medium could successfully induce adventitial roots,and the number and length of adventitious roots induced by M_(2) were significantly higher than the control.[Conclusion]The effects of 8 brands of powder MS medium were significant different in the tissue culture of E.purpurea and A.roxburghii,and except for the medium codes M_(5),M_(6),and M_(8),other brands of powder MS medium are not suitable for tissue culture of E.purpurea.However,except for M_(4),all 7 other brands of powder MS medium are suitable for A.roxburghi.
