摘要
尿胰蛋白酶抑制剂(Urinary Trypsin Inhibitor,UTI)是从人类尿液中分离纯化得到的一种具有丝氨酸蛋白酶抑制活性的硫酸软骨素蛋白聚糖,其对胰蛋白酶、透明质酸酶、粒细胞弹性蛋白酶、纤溶酶等蛋白酶均具有良好抑制作用,被广泛用于临床治疗急性胰腺炎和作为急性循环衰竭的抢救辅助用药。本文主要综述了尿胰蛋白酶抑制剂的来源、结构、生理功能、传统分离纯化工艺以及基因工程技术生产新方法等,旨在为尿胰蛋白酶抑制剂的高质量生产以及应用提供参考。
Urinary trypsin inhibitor(UTI),isolated and purified from human urine,is a small chondroitin sulfate proteoglycan with Ser-proteinase inhibitory activity.UTI have good inhibitory effects on proteases such as trypsin,hyaluronidase,neutrophil elastase,and plasmin,and have been widely used in clinical treatment of acute pancreatitis,and rescue medication for acute circulatory failure.In present review,the sources,structure,physiological functions,conventional separation and purification processes,and the emerging trends in genetic engineering methods of UTI are summarized,in order to provide consultation for the high-quality production and application of UTI.
