组织RP11-388M20.1水平对胃癌根治术患者预后的评估价值

Prognostic value of tissue RP11-388M20.1 level in patients with gastric cancer undergoing radical gastrectomy

目的 探讨胃癌组织中长链非编码RNA RP11-388M20.1水平及其评估胃癌根治术患者预后的价值。方法 选取2018年1月至2020年12月于本院行胃癌根治术的患者103例,采用实时荧光定量PCR检测RP11-388M20.1在胃癌组织及对应癌旁组织中的表达差异,分析RP11-388M20.1水平与胃癌患者临床病理特征的关系,采用Kaplan-Meier生存曲线并基于Cox比例风险模型分析RP11-388M20.1在胃癌根治术患者预后预测中的价值。结果 103例胃癌组织的RP11-388M20.1水平为5.013±0.184,高于癌旁组织的1.471±0.056(t=18.399,P<0.001)。RP11-388M20.1在胃癌组织中的表达显著与TNM分期(P=0.005)、肿瘤大小(P=0.038)、分化程度(P=0.002)、T分期(P=0.029)和癌胚抗原状态(P=0.022)有关。单因素分析结果显示RP11-388M20.1表达与胃癌预后有关,其中RP11-388M20.1低表达者的中位无病生存期未达,优于高表达者的36个月(HR=2.936,95%CI:1.808~4.768,P<0.001);多因素分析显示RP11-388M20.1高表达是胃癌预后不良的危险因素(HR=2.076,95%CI:1.070~4.026,P=0.031)。结论 RP11-388M20.1在胃癌组织中是一种过表达的长链非编码RNA,且与胃癌进展和不良预后有关,有作为胃癌诊治靶点的潜能。

Objective To investigate the level of long non-coding RNA RP11-388M20.1 in gastric cancer tissues and its value in evaluating the prognosis of patients undergoing radical gastrectomy.Methods One hundred and three patients who underwent radical gastrectomy for gastric cancer in our hospital from January 2018 to December 2020 were selected.Rea-time quantitative PCR was used to detect the differential expression of RP11-388M20.1 between gastric cancer tissues and corresponding adjacent tissues.Relationship of RP11-388M20.1 level with clinical pathological characteristics of gastric cancer patients were analyzed.Kaplan-Meier survival curve and Cox proportional risk model were applied to analyze the value of RP11-388M20.1 in predicting the prognosis of gastric cancer patients undergoing radical surgery.Results The RP11-388M20.1 level in 103 gastric cancer tissues was 5.013±0.184,higher than the 1.471±0.056 in adjacent tissues(t=18.399,P<0.001).The expression of RP11-388M20.1 in gastric cancer tissues was significantly associated with TNM staging(P=0.005),tumor size(P=0.038),differentiation degree(P=0.002),T staging(P=0.029),and carcinoembryonic antigen status(P=0.022).Results of univariate analysis showed that the expression of RP11-388M20.1 was related to the prognosis of gastric cancer.Among them,the median disease-free survival of patients with low expression of RP11-388M20.1 was unreached,better than 36 months of those with high expression(HR=2.936,95%CI:1.808-4.768,P<0.001);Multivariate analysis showed that high expression of RP11-388M20.1 was a risk factor for poor prognosis in gastric cancer(HR=2.076,95%CI:1.070-4.026,P=0.031).Conclusion RP11-388M20.1 is an overexpressed long non-coding RNA in gastric cancer tissues,which is associated with gastric cancer progression and poor prognosis,and has the potential to serve as a diagnostic and therapeutic target for gastric cancer.
