摘要
目的:探讨改进后的康复器械电刺激皮层对杏仁核部分损伤引起小鼠焦虑样行为能力缺失的干预机制。方法:对脑电仿生电刺激治疗仪探头改造,使其适用于C57小鼠。选取C57小鼠作为研究对象。先对小鼠进行旷场实验、高架十字迷宫行为实验,并在浅麻醉状态下测试小鼠的心率(心率测试);然后物理损伤小鼠双侧基底外侧杏仁核(前后方向=2.24,中外侧方向=±2.90,背腹方向=4.75),重复旷场实验、高架十字迷宫行为学实验和心率测试;再用电刺激小鼠皮层,重复旷场实验、高架十字迷宫行为学实验和心率测试。结果:损伤小鼠双侧基底外侧杏仁核导致焦虑样行为能力显著下降(P<0.05,单因素方差分析),伴随着心率下降(P<0.05,单因素方差分析);电刺激小鼠皮层后,小鼠焦虑样行为能力有所上升(P<0.05,单因素方差分析),伴随着心率上升(P<0.05,单因素方差分析)。结论:损伤基底外侧杏仁核导致的小鼠焦虑样行为程度的下降,伴随心率下降;电刺激皮层可以提升小鼠焦虑样行为能力,伴随心率上升。
Objective:To investigate the intervention mechanism of improved rehabilitation equipment electrical stimulation of cortical on anxiety like behaviour deficit induced by partial injury of amygdala in mice.Methods:Transform the probe of the EEG biomimetic electrical stimulator treatment device to make it suitable for C57 mice.C57 mice were selected as subjects.First,performed open field test(OFT)and the elevated plus maze(EPM)on mice;And measured the heart rate of mice under light anesthesia(HR test).Second,injured the bilateral BLA(AP=2.24,ML=±2.90,DV=4.75)of mice,and repeated OFT,EPM and HR test.Then electrical stimulated the mice cortexes,and repeated OFT,EPM and HR test.Results:Partial lesion of bilateral amygdala in mice resulted in significant decrease of anxiety like behavior ability(P<0.05,one way ANOVA),accompanied by a decrease in heart rate(P<0.05,one way ANOVA);After stimulated on cortexes of mice,the anxiety like behaviors ability of mice were increased(P<0.05,one way ANOVA),accompanied by an increase in heart rate(P<0.05,one way ANOVA).Conclusion:BLA injury caused the decrease of anxiety like behavior levels in mice,accompanied with decrease in HR.Electrical stimulation of the cortex can improve the levels of anxiety like behavior in mice,accompanied with increase in HR.
