脑缺血再灌注对腹部手术大鼠海马神经元和小胶质细胞白细胞介素-1R1表达的影响

Effects of cerebral ischemia-reperfusion on hippocampal neurons and microglias interleukin-1R1 expression in rats undergoing abdominal surgery

摘要 目的探讨脑缺血再灌注损伤对腹部手术大鼠海马神经元和小胶质细胞白细胞介素-1R1(IL-1R1)表达的影响。方法24只8~10月龄雄性SD大鼠分为3组:假手术组(Sham)、腹部手术组(Surg)、腹部手术+脑缺血再灌注组(Surg+IR)。分别予以只麻醉无手术、麻醉下腹部手术、麻醉下腹部手术同时颈总动脉结扎10 min松开这3种处理。术后48 h,大鼠深麻醉下心脏灌流剥离全脑,分离海马,固定切片后,进行免疫荧光染色和共聚焦显微镜观察拍照,采用Image J计数IL-1R1阳性的神经元和小胶质细胞个数。组间比较采用单因素方差分析(ANOVA)。结果Sham组、Surg组和Surg+IR组大鼠海马DG区神经元的IL-1R1阳性染色数目分别为(37.50±5.63)、(42.62±5.73)、(86.00±6.35)个;与Sham组比较,Surg组阳性染色数目差异无统计学意义(F=3.25,P>0.05),Surg+IR组阳性染色数目显著高于Sham组(F=261.36,P<0.05);Surg+IR组阳性染色数目亦显著高于Surg组(F=205.83,P<0.05)。Sham组、Surg组和Surg+IR组大鼠海马DG区小胶质细胞的IL-1R1阳性染色数目分别为(24.00±3.55)、(24.87±3.80)、(103.37±9.35)个;与Sham组比较,Surg组阳性染色数目差异无统计学意义(F=0.23,P>0.05),Surg+IR组阳性染色数目显著高于Sham组(F=504.12,P<0.05);Surg+IR组阳性染色数目亦显著高于Surg组(F=484.16,P<0.05)。结论脑缺血再灌注损伤上调腹部手术大鼠海马神经元和小胶质细胞IL-1R1表达水平。 Objective To investigate the effects of cerebral ischemia-reperfusion injury(CIRI)on interleukin-1R1(IL-1R1)expression in hippocampal neurons and microglia of rats undergoing abdominal surgery.Methods Totally,2418-month-old SD rats were randomly divided into three groups(n=8 each):sham-operated(Sham)group,abdominal surgery(Surg)group,abdominal surgery+CIRI(Surg+IR)group,which were given general anesthesia without surgical operation,general anesthesia and abdominal surgical treatment,general anesthesia and abdominal surgery and meanwhile common carotid ligation for 10 min,respectively.Then the rats were sacrificed by heart perfusion and the whole brain was stripped 48 h after operation.The hippocampi of the rats were extracted for immunofluorescence staining and confocal observation.The number of IL-1R1 positive neurons and microglias were calculated using Image J.One way analysis of variance(ANOVA)was used for comparison between groups.Results The number of IL-1R1 positive neurons in hippocampal dentate gyrus of Sham group,Surg group and Surg+IR group was 37.50±5.63,42.62±5.73,86.00±6.35,respectively.No significant difference in the number of IL-1R1 positive neurons was observed between Sham group and Surg group(F=3.25,P>0.05),while the number in Surg+IR group was significantly greater than that in Sham group(F=261.36,P<0.05).The number of IL-1R1 positive neurons in Surg+IR group was also significant greater than in Surg group(F=205.83,P<0.05).The number of IL-1R1 positive microglias in hippocampal dentate gyrus of Sham group,Surg group and Surg+IR group was 24.00±3.55,24.87±3.80,103.37±9.35,respectively.No significant difference in the number of IL-1R1 positive microglias was observed between Sham group and Surg group(F=0.23,P>0.05),while the number in Surg+IR group was significantly greater than that in Sham group(F=504.12,P<0.05).The number of IL-1R1 positive microglias in Surg+IR group was also significant greater than in Surg group,(F=484.16,P<0.05).Conclusion CIRI upregulates IL-1R1 expression levels in hippocampal neurons and microglia of rats undergoing abdominal surgery.
