摘要
本文构建了4T1细胞适配体编程策略以精确控制微孔板上4T1细胞的捕获和释放。同时开发了一种具有良好过氧化物酶(POD)活性的纳米催化剂PB/PM/HRP/Apt,其可在H2O2存在下,催化显色剂3,3’,5,5’-四甲基联苯胺(TMB)氧化生成蓝色氧化物oxTMB。在优化条件下,利用比色法对4T1细胞进行定量分析。结果显示,吸光度与4T1细胞浓度在1×10^(4)~1×10^(6)个/mL范围内呈良好的线性关系。本文提出的治疗策略,操作简单明了,且显著提高了对4T1细胞的捕获和定量分析能力。
A 4T1 cell aptamer programming strategy was constructed to accurately control the capture and release of 4T1 cells on the microplate.In addition,a nano-catalyst PB/PM/HRP/Apt,with good peroxidase(POD)activity,was developed.It can catalyze color developing agent 3,3',5,5'-tetramethylbenzidine(TMB)to generate blue oxide oxTMB quickly in the presence of H2O2.Under the optimal conditions,the quantitative analysis of 4T1 cells was achieved using colorimetric method,and the absorbance showed a good linear relationship with the concentration of 4T1 cells in the range of 1×10^(4)-1×10^(6) cells/mL.This developed therapeutic strategy could be operated simply,and could significantly improve the ability of capturing and quantitatively analyzing for 4T1 cells.
