桑杜口服液对环磷酰胺致鸡免疫抑制的调节作用

摘要 为研究桑杜口服液对鸡免疫抑制的调节作用,试验采用环磷酰胺(40 mg/kg)建立鸡免疫抑制模型,并给予鸡传染性法氏囊病活疫苗(B87株)免疫和不同剂量桑杜口服液(分别为4.8 g/kg、9.6 g/kg和19.2 g/kg)处理,由此将120只13日龄雏鸡随机均分为6组,通过抗体水平检测、组织切片和RT-qPCR等技术评估不同剂量桑杜口服液对环磷酰胺诱导免疫抑制的调节作用。结果显示:9.6 g/kg和19.2 g/kg桑杜口服液预处理可有效缓解环磷酰胺造成的B87疫苗抗体生成减少、法氏囊指数下降和法氏囊滤泡损伤现象。此外,在鸡传染性法氏囊病活疫苗(B87株)免疫的基础上,环磷酰胺处理显著降低白细胞介素-4(IL-4)、白细胞介素-10(IL-10)和γ-干扰素(IFN-γ)mRNA表达量(P<0.05),但未对白细胞介素-2(IL-2)表达量产生显著影响(P>0.05),而9.6 g/kg桑杜口服液处理显著降低IL-2 mRNA表达量(P<0.05),19.2 g/kg桑杜口服液可显著升高IFN-γ mRNA表达量(P<0.05)。此外,9.6 g/kg和19.2 g/kg桑杜口服液可显著拮抗环磷酰胺对IL-4和IL-10的抑制作用(P<0.05)。研究表明,19.2 g/kg桑杜口服液可以缓解环磷酰胺造成的鸡的免疫抑制。 In order to study the regulation effect of Sangdu oral liquid on immune suppression in chicken,the chicken immu-nosuppression model was established by cyclophosphamide(40 mg/kg),and the chickens were immunized with infectious bursal disease vaccine(B87 strain)and treated with different dose of Sangdu oral liquid(4.8 g/kg,9.6 g/kg and 19.2 g/kg,respectively),on this basis,12013-day-age chickens were randomly divided into 6 groups.The regulatory effect of different doses of Sangdu oral liquid on cyclophosphamide induced immunosuppression was evaluated by antibody level detection,tissue section and RT qPCR.The results showed that pretreatment of Sangdu oral liquid at the doses of 9.6 g/kg and 19.2 g/kg could effectively alleviate the reduction of B87 vaccine antibody production,the decrease of Fabricius vesicle index and the damage of Fabricius vesicles caused by cyclophosphamide.In addition,on the basis of B87 strain vaccine immunization,cyclophosphamide treatment signifi-cantly decreased the mRNA expression of interleukin-4(IL-4),interleukin-10(IL-10)and interferonγ(IFN-γ)(P<0.05),but had no significant effect on the expression of interleukin-2(IL-2)(P<0.05),while 9.6 g/kg Sangdu oral liquid treatment significant-ly decreased the mRNA expression of IL-2(P<0.05),19.2 g/kg Sangdu oral liquid significantly increased the mRNA expression of IFN-γ(P<0.05).In addition,9.6 g/kg and 19.2 g/kg Sangdu oral liquid could significantly antagonize the inhibitory effect of cyclo-phosphamide on IL-4 and IL-10(P<0.05).The above results indicated that 19.2 g/kg Sangdu oral liquid could inhibit the immuno-suppression caused by cyclophosphamide in chicken.
