基于响应面优化陈皮黄酮类有效成分的提取工艺

Optimization of Extraction Technology of Flavonoids from Dried Tangerine Peel Based on Response Surface

摘要 以陈皮为研究原料,以陈皮中的总黄酮提取率为响应值,采用单因素以及响应面实验优化索氏提取陈皮黄酮类有效成分的工艺。考察液料比、粉碎粒度、提取时间和乙醇体积分数对陈皮总黄酮提取率的影响,并采用Box-Behnken实验设计的四因素三水平响应面分析方法优化工艺。结果表明陈皮黄酮类有效成分提取的最佳工艺条件是:液料比为20∶1(mL/g);粉碎粒度为100目;提取时间为2.5 h;乙醇体积分数为35%。此条件下陈皮总黄酮的提取率为1.65%,预测提取率为1.68%,与实际值非常吻合。在此最佳工艺条件下提取的陈皮浸膏经乙醚萃取后,得到的乙醚萃取物进一步用气相色谱-质谱(gas chromatogrphy-mass spectrometry, GC-MS)法分析,共鉴定出27种挥发性成分,其中主要成分是多甲氧基黄酮类化合物。 Using dried tangerine peel as row material,and the extraction rate of total flavonoids from tangerine peel as the response value,the process of Soxhlet extraction of flavonoids from tangerine peel was optimized by single factor test and response surface test.The effects of extraction time,ethanol volume fraction,liquid-solid ratio and crushing particle size on the extraction rate of total flavonoids from tangerine peel were examined,and the four factor and three-level response surface analysis method designed by Box Behnken experiment was used to optimize the process.The results showed that the optimum extraction conditions of flavonoids from tangerine peel were as follows:extraction time was 2.5 h;ethanol volume fraction was 35%,liquid-solid ratio was 20∶1(mL/g);crushing particle size was 100 mesh.And the extraction rate of total flavonoids from tangerine peel was 1.65%.The predicted extraction amount was 1.68%,which was very consistent with the actual value.After the extract of tangerine peel was extracted by ether with the optimum conditions,the ether extract was further analyzed by GC-MS.A total of 27 volatile components were identified,and the main components were polymethoxy flavonoids.
