新型合成阿片类物质美托尼秦的定性检验

摘要 本文针对在中国未见报道的新型合成阿片类物质美托尼秦,建立其气相色谱-质谱(GC-MS)、超高效液相色谱-四极杆/静电场轨道阱高分辨质谱联用(UPLC-Q-Exactive Orbitrap-MS)定性检验方法。使用甲醇提取未知样品,取上清液,分别采用GC-MS和UPLC-Q-Exactive Orbitrap-MS进行分析,使用异丙托尼秦(isotonitazene)进行同步对照分析。GC-MS检测结果表明,保留时间为15.167 min的未知组分的质谱主要特征碎片离子峰有m/z 86.1(基峰)、58.05、121.1、205.05、236.05和248.1。UPLC-Q-Exactive Orbitrap-MS检测结果表明,保留时间为7.35 min的未知组分准分子离子峰为m/z 383.2077,二级质谱主要特征碎片离子有m/z 310.1182、264.1252、121.0649、100.1121和72.0808。基于缺少对照品的现状,经SWGDRUG和Cayman谱库检索、与已报道文献中信息比对及对其高分辨质谱数据的解析,实现了目标物确认,鉴定为美托尼秦(metonitazene)。该方法具有分析简便、快速的特点,有很好的应用前景,可以用于实际案件的检测。 New synthetic opioids are on the growing list of illicit drugs and pose a serious threat to human health.Some examples of NSOs include AP 237,piperidine,bromomorphine and a variety of 2-benzyl benzimidazole opiates.2-benzyl benzimidazole opioids,also known as“Nitazenes”,many of which are regulated by the United Nations Commission on Narcotic Drugs,have become internationally prevalent in recent years.Metonitazene is an emerging potent synthetic opioid thatfirst appeared in the recreational drug supply in mid-2020 and began to surge by the end of the decade,raising increasing public health concerns.In this study,we presented a method for the identification of a novel synthetic opioid metonitazene seized for the first time in China by gas chromatography-mass spectrometry(GC-MS)and ultra-high performance liquid chromatography-quadrupole/electrostaticfield Orbitrap-MS(UPLC-Q-Exactive Orbitrap-MS).The unknown samples were extracted by methanol,and the supernatants were analyzed by GC-MS and UPLC-Q-Exactive Orbitrap-MS respectively.Isotonitazene was analyzed as a contrast.By GC-MS detection,the main characteristic ion peaks of the mass spectral fragments of unknown component with retention time of 15.167 min were m/z 86.1(base peak),58.05,121.1,205.05,236.05 and 248.1.UPLC-Q-Exactive Orbitrap-MS detection showed that the excimer ion peak of unknown component with retention time of 7.35 min was m/z 383.2077.The main ions of secondary mass spectrum were m/z 310.1182,264.1252,121.0649,100.1121 and 72.0808.Due to the lack of reference substance,the target was identified as metonitazene by retrieval of SWGDRUG and Cayman database,comparison of information in reported literatures and analysis of high-resolution mass spectrometry data.The method is simple,rapid and has good application prospect.It can be used in real case detection.
