基于HPLC含量测定评价鸡血藤粉碎后纤维样物质的利用价值

Evaluation of utilization value of fiber-like substance after crushing Spatholobus Suberectus based on HPLC content determination

摘要 目的建立HPLC梯度洗脱法测定鸡血藤粉碎后细粉和纤维样物质中儿茶素与表儿茶素的含量,为评价纤维样物质利用价值提供依据。方法采用RD-C18色谱柱(4.6 mm×250 mm,5μm),以0.1%磷酸水溶液为流动相A,乙腈为流动相B,实行梯度洗脱模式,流速为0.8 mL/min,检测波长260 nm,柱温为30℃。采用SPSS 26.0软件t检验分析儿茶素、表儿茶素在细粉和纤维样物质中的含量差异。结果儿茶素和表儿茶素检测质量浓度线性范围分别为0.0454~0.2270 mg/mL(R^(2)=0.9997)、0.0622~0.3110 mg/mL(R^(2)=0.9993),平均加样回收率分别为102.31%、98.70%。t检验结果显示,鸡血藤粉碎后细粉和纤维样物质中儿茶素、表儿茶素含量差异均没有统计学意义(P>0.05)。结论建立的方法简便、灵敏、准确度高,可为评价鸡血藤粉碎后产生的纤维样物质利用价值提供依据。纤维样物质和细粉在主要活性成分上具有一定的质量等同性,故不建议丢弃纤维样物质。 Objective To establish the HPLC method with gradient elution for the content determination of catechin and epicatechin in fine powder and fiber-like substance after crushing Spatholobus Suberectus,so as to provide reference for evaluation of utilization value of fiber-like substance.Methods The gradient elution was performed on a RD-C18 column(4.6 mm×250 mm,5μm)with 0.1% phosphoric acid aqueous solution as the mobile phase A and acetonitrile as the mobile phase B.The flow rate was 0.8 mL/min and the wavelength was 260 nm.The column temperature was 30℃.The content difference of catechin and epicatechin in fine powder and fiber-like substance was analyzed by t-test with SPSS 26.0 software.Results The linear ranges of catechin and epicatechin were 0.0454-0.2270 mg/mL(R^(2)=0.9997)and 0.0622-0.3110 mg/mL(R^(2)=0.9993),respectively.The average recoveries were 102.31%and 98.70%,respectively.The results of t-test showed that there was no significant difference between the contents of catechin and epicatechin in fine powder and fiber-like substance after crushing Spatholobus Suberectus(P>0.05).Conclusion The established method is simple,sensitive and accurate,which can provide a reference for evaluating the utilization value of fiber-like substance after crushing Spatholobus Suberectus.The fine powder and the fiber-like substance have certain quality equivalence in main active components,so it is not recommended to discard fiber-like substances.
