摘要
目的以青翘为原料,比较不同提取工艺的提取效果,并重点分析不同青翘提取物中连翘苷的含量。方法采取控制变量法原则,分别使用加热回流提取法、超声波提取法、微波萃取法等提取工艺进行提取;采用WelchUltimateC18(4.6×250mm,5μm)色谱柱,流动相为乙腈(B)—水(A),V(乙腈):V(水)=30:70;流速为1.0mL·min^(-1),柱温30℃,检测波长为277nm,进样量为10μL。结果通过比较分析,选择加热回流法进行提取具有最佳的提取效果,青翘提取物经AB-8型树脂纯化后使用HPLC进行定量检测,其连翘苷含量为1.386‰。结论该提取方法简便、提取效果充分,HPLC检测方法灵敏、准确、重现性好,可作为青翘中连翘苷含量测定和质量控制的依据。
Objective in this study,Forsythia suspensa was used as raw material to compare the extraction effects of different extraction processes,and the content of forsythin in different Forsythia suspensa extracts was mainly analyzed.Methods According to the principle of controlled variable method,heating reflux extraction,ultrasonic extraction and microwave extraction were used,respectively;Welch ultimate C18(4.6×250 mm,5μm)chromatographic column was used.The mobile phase was acetonitrile(b)-water(a),V(acetonitrile):V(water)=30:70;The flow rate was 1.0 mL·min^(-1),the column temperature was 30℃,the detection wavelength was 277 nm,and the injection volume was 10μL.Results Through comparative analysis,the best extraction effect was obtained by heating reflux method.The extract of Forsythia suspensa was purified by AB-8 resin and quantitatively detected by HPLC.The content of forsythin was 1.386‰.Conclusion The extraction method is simple and effective.The HPLC detection method is sensitive,accurate and reproducible,which can be used as the basis for the content determination and quality control of forsythin in Forsythia suspensa.
