响应面优化“漯紫薯4”花色苷提取工艺及其抗氧化活性研究

Optimization of Extraction Process for“Luozishu 4”Anthocyanins by Response Surface Methodology and Their Antioxidant Activity

摘要 以“漯紫薯4”为材料,在单因素试验的基础上,采用响应面法对紫薯花色苷的提取工艺进行了优化试验,并对紫薯花色苷的抗氧化活性进行了研究。结果表明,3个单因素对花色苷得率影响大小的顺序为盐酸体积分数>料液比>提取时间,紫薯花色苷最佳提取工艺参数为盐酸体积分数0.75%、料液比1∶20(g/mL)、提取时间20 min。在最佳提取工艺条件下,花色苷的平均得率达到108.55 mg/100 g,RSD为0.05%,与理论值(108.84 mg/100 g)的相对误差为0.27%。抗氧化活性试验研究表明,当紫薯花色苷和维生素C的浓度相同时,其各自的抗氧化活性的变化趋势一致,同时,紫薯花色苷对DPPH具有较好的清除能力且还原力也较好,紫薯花色苷的抗氧化能力强于维生素C。 Using“Luozishu 4”as experimental material,the extraction process of purple sweet potato anthocyanin was optimized by response surface methodology on the basis of single factor experiment,and the antioxidant activity of purple sweet potato anthocyanin was studied.The results showed that the order of the influence of three single factors on the yield of anthocyanin was C(HCl volume fraction)>B(material-liquid)>A(extraction time).The optimal extraction parameters of purple sweet potato anthocyanin were HCl volume fraction 0.75%,material-liquid ratio 1∶20(g/mL),and extraction time 20 min.Under the optimal extraction conditions,the average yield of anthocyanin reached 108.55 mg/100 g,with RSD of 0.05%,and the relative error with the theoretical value(108.84 mg/100 g)was 0.27%.The antioxidant activity test showed that purple sweet potato anthocyanin had good scavenging ability to DPPH and good reducing ability.When the concentrations of purple sweet potato anthocyanin and vitamin C were the same,their antioxidant change trend were the same,and the antioxidant ability of purple sweet potato anthocyanin was stronger than that of vitamin C.
