日粮中添加姜黄粉对黄羽肉鸡抗氧化和脂质代谢的影响

Effects of dietary supplementation with turmeric powder on antioxidant and lipid metabolism in yellow-feathered broilers

摘要 阐明日粮中添加不同水平姜黄粉对黄羽肉鸡抗氧化功能、脂质代谢等方面的影响,可为姜黄粉在黄羽肉鸡生产养殖中的应用提供理论基础.选取1日龄健康雄性快速型黄羽肉鸡320羽,随机分为4组,每组8个重复,每个重复10羽.对照组:饲喂基础饲粮;姜黄素组:基础饲粮中添加150 mg/kg姜黄素;0.5%姜黄粉组:在基础饲粮中添加5 g/kg姜黄粉;1%姜黄粉组:在基础饲粮中添加10 g/kg姜黄粉(姜黄粉中的姜黄素含量约3%).试验期70 d,分别于第28天、第70天测定肉鸡血清和肝的抗氧化、脂质代谢等相关指标,并通过RT-qPCR检测肝抗氧化和脂质代谢相关基因表达水平.结果显示:姜黄粉和姜黄素均显著性提高了黄羽肉鸡血清和肝的SOD、GSH-Px等抗氧化酶活性(P<0.05),并且一定程度上改善了肉鸡血脂和肝脂代谢水平.同时,姜黄粉和姜黄素均调节了黄羽肉鸡肝Keap1、Nrf2、HO-1等抗氧化相关基因以及FAS、ACC、HSL、PPARα等脂质代谢相关基因的表达.本研究表明姜黄粉可以有效调节黄羽肉鸡肝抗氧化和脂质代谢相关基因表达,提高黄羽肉鸡血清和肝抗氧化能力,调节黄羽肉鸡脂质代谢水平;且整体上姜黄粉添加剂量为1%时效果更显著. This study was conducted to investigate the regulatory effects of dietary supplementation with turmeric powder on redox homeostasis and lipid metabolism in yellow-feathered broilers,aiming to provide a theoretical basis for the application of turmeric powder in the production of yellow-feathered broilers.Three hundred and twenty one-day-old healthy male fast-type yellow-feathered broilers were randomly divided into four groups with eight replicates per group and 10 broilers per replicate.The control group was fed a basal diet;the curcumin group incorporated 150 mg/kg curcumin into the basal diet;the 0.5%turmeric powder group incorporated 5 g/kg turmeric powder into the basal diet;the 1%turmeric powder group incorporated 10 g/kg turmeric powder into the basal diet.The experiment lasted for 70 days.Antioxidant and lipid metabolism indices in the serum and livers of broilers were measured on days 28 and 70,respectively,and the expression levels of genes related to antioxidant and lipid metabolism in the liver were detected using qRT-PCR.Both turmeric powder and curcumin significantly increased antioxidant enzyme activity,including SOD and GSH-Px,in both serum and liver(P<0.05),and serum and liver lipid metabolism of broilers improved to a certain extent.Expression of antioxidantrelated genes including Keap1,Nrf2 and HO-1,and lipid metabolism-related genes including FAS,ACC,HSL,and PPARαin the liver of yellow-feathered broilers were effectively regulated by both curcumin and turmeric powder.In conclusion,dietary supplementation with turmeric powder can effectively regulate the expression of genes that regulate antioxidant and lipid metabolism in the livers of yellow-feathered broilers,improve antioxidant capacity in the serum and liver,and regulate lipid metabolism in yellow-feathered broilers.The effect was most significant with 1%supplemental turmeric powder.
