霍山石斛黄酮的闪式提取工艺优化及其抗疲劳功能研究

Optimization of Flash Extraction Process for Flavonoids from Dendrobium huoshanense and Its Anti-Fatigue Function

摘要 目的:优化霍山石斛黄酮的提取工艺并研究其抗疲劳特性。方法:利用单因素和响应面试验优化霍山石斛黄酮闪式提取工艺,并通过小鼠负重游泳试验,测定血乳酸、血尿素氮、肝糖原等指标评估其抗疲劳能力。结果:在提取电压112 V、提取时间54 s、乙醇浓度52%、料液比1:34(g/mL)的条件下霍山石斛黄酮的提取工艺效果最佳,此时霍山石斛黄酮提取量为(1.29±0.0697)mg/g。动物试验表明,霍山石斛黄酮能提高小鼠负重游泳时间,降低血乳酸和血尿素氮含量(P<0.05),提升肝糖原含量(P>0.05)。结论:霍山石斛黄酮具有一定的抗疲劳能力,该结果可为其进一步研究和开发提供理论依据。 Objective:Extraction process of flavonoids from Dendrobium huoshanense was optimized and its anti-fatigue properties were studied.Method:The single-factor tests and response surface analysis was adopted to optimize the flash extraction process of Dendrobium huoshanense flavonoids.The anti-fatigue capacity was evaluated through a weight-loaded swimming test in mice,measuring indicators such as blood lactic acid,blood urea nitrogen,and hepatic glycogen.Result:The optimized processing conditions were determined to include an extraction voltage 112 V,extraction time 54 s,ethanol concentration 52%and substrate-to-solvent ratio 1:34(g/mL).Under these conditions,the extraction yield of flavonoids from Dendrobium huoshanense was(1.29±0.0697)mg/g.Animal experiments showed that flavonoids from Dendrobium huoshanense could increase the swimming time of weight-loaded mice,reduce blood lactic acid and blood urea nitrogen levels(P<0.05),and increase liver glycogen levels(P>0.05).Conclusion Dendrobium huoshanense flavonoids could improve the anti-fatigue ability on mice,which provided a basis for the further research and development of Dendrobium huoshanense flavonoids.
