聚羟基脂肪酸酯的萃取精制工艺研究

摘要 为推动中国新型生物基高分子材料的全面发展,促进生物基材料PHA的高值化应用,通过筛选萃取剂种类和用量、预处理试剂种类和用量以及预处理时间,确定了PHA样品最佳萃取工艺参数,并显著提高了PHA产品的质量和回收效率。结果表明,最佳预处理试剂为次氯酸钠,萃取剂为三氯甲烷,2种试剂的使用量分别是PHA样品质量的14倍和19倍,预处理时间1.5 h时发酵效果最好,最终纯度和回收率分别为98.35%和89.60%。该研究以三氯甲烷和次氯酸钠萃取PHA,显著提升PHA纯度和回收率,并简化提取工艺,为推进生物基材料PHA的下游分离提取工作提供有力的理论支持。 In order to promote the comprehensive development of new biopolymer materials in China and the highefficiency application of PHA biomaterial,the optimal extraction process parameters of PHA samples were determined,and the quality and extraction efficiency of PHA products were significantly improved by selecting the type and amount of extractant,the type and amount of pretreatment reagent and the pretreatment time.The results showed that the optimal pretreatment reagent was sodium hypochlorite and the extractant was trichloromethane,and the amounts of both reagents were 14 and 19 times of the mass of PHA sample,respectively,and the best fermentation effect was achieved at a pretreatment time of 1.5 h.The final purity and recovery were 98.35%and 89.60%,respectively.In this study,PHA extraction was carried out using trichloromethane and sodium hypochlorite,which significantly improved the purity and recovery of PHA and simplified the extraction process,providing strong theoretical support in promoting the subsequent separation and recovery of PHA from biomaterials.
