摘要 目的 鉴定五倍子化学成分并探究其舒张血管的作用机制。方法 建立五倍子UNIFI数据库。采用超高效液相色谱-四级杆-飞行时间-质谱联用技术(UPLC-Q-TOF-MS^(E))与UNIFI数据处理平台,鉴定五倍子中的化学成分。用大鼠胸主动脉环实验探究五倍子提取物舒张血管的作用和其有效的物质成分发挥作用的机制。结果 超高效液相色谱串联四级杆飞行时间质谱技术结合UNIFI数据库快速分析鉴定五倍子化学成分60种。五倍子提取物可能通过促进血管内皮一氧化氮合酶的活性;在药理通路中,它可能影响平滑肌细胞内Ca2+的释放和外钙内流,从而发挥作用。结论 本研究首次采用UPLC-Q-TOF-MS^(E)结合UNIFI数据库对五倍子提取物进行鉴定。从五倍子提取物中鉴定出60种化合物。酚酸类占比较大,其药理作用可能是酚酸类化合物通过内皮依赖和内皮非依赖途径激活一氧化氮合酶发挥舒张血管的作用。药理通路其可能为通过促进平滑肌细胞内钙释放和外钙内流发挥作用。 Objective To explore the vasodilation effect of gallnut and the mechanism of its effective components.Methods The UNIFI database of gallnut was established for the chemical constituents of gallnut identification by UPLC-Q-TOF-Mass system and UNIFI data processing platform.The vasodilation effect of Galla chinensis extract and its possible mechanism via the experiment of rat thoracic aorta ring were explored.Results UPLC-QTOF-MSE combined with UNIFI database was used to rapidly analyze and identify 60 chemicals of Galla chinensis,and most of them was phenolic acid.Rat thoracic aorta ring experiment interfered with extract of Galla chinensis might reveal the relax blood vessels effect via endothelium-dependent and endothelium-independent pathways.It might play a role through vascular endothelial nitric oxide synthase.Pharmacological of its pathway might play a role by promoting intracellular calcium release and external calcium influx in smooth muscle cells.Conclusion This study is the first time to identify gallnut extract by UPLC-Q-TOF-MSE combined with UNIFI database and 60 compounds were identified from the extract of gallnut.Phenolic acids account for a large proportion,and their pharmacological effects may be that phenolic acids activate nitric oxide synthase through endothelium-dependent and endothelium-independent pathways to relax blood vessels.Pharmacological pathway may play a role by promoting intracellular calcium release and external calcium influx in smooth muscle cells.
