黄芩苷对扑热息痛诱导的小鼠肝脏氧化应激和凋亡的影响

摘要 为探究黄芩苷(BA)对扑热息痛(APAP)诱导急性肝损伤的保护作用,并对其机制进行初步探讨。试验将小鼠随机分为空白对照组(CON组)、黄芩苷对照组(BA组)、APAP模型对照组(APAP组)、黄芩苷低剂量组(APAP+L-BA组)和黄芩苷高剂量组(APAP+H-BA组)。H&E染色制作病理组织切片,观察肝脏病理学变化,检测血清ALT、AST水平,用试剂盒测定肝脏GSH、GSH-Px、MDA水平,Western blot测定Bax和Bcl-2的蛋白表达水平。结果表明:与CON组相比,APAP组血清中ALT、AST水平上升,肝组织GSH和GSH-Px的含量下降,MDA水平上升,Bax的表达增加,Bcl-2的表达减少,且H&E染色结果显示肝脏明显损伤;与APAP组相比,APAP+L-BA组和APAP+H-BA组血清ALT、AST、MDA水平降低,GSH和GSH-Px的含量升高,Bax的蛋白表达水平降低,Bcl-2的蛋白表达增加,且H&E染色结果显示肝脏受损情况明显减轻。综上,BA对APAP诱导的小鼠急性肝损伤具有保护作用,其机制可能与减轻小鼠肝脏内氧化应激引起的自由基-抗氧化酶失衡和调节凋亡相关蛋白Bax、Bcl-2表达有关。 To explore the baicalin(BA)on acute liver injury induced by acetaminoPhen(APAP)protection,and preliminarily discuss its mechanism,the mice were randomly divided into blank control group(CON group),baicalin control group(BA group),APAP model control group(APAP group),baicalin low-dose group(APAP+L-BA group)and baicalin high-dose group(APAP+H-BA group).Pathological tissue sections was made by H&E staining,liver pathological changes were observed,serum ALT and AST levels were detected,liver GSH,CSH-Px and MDA levels were measured by kit,and protein expression levels of Bax and Bcl-2 were tested by Western blot.The test results showed that compared with the CON group,ALT increased,CSH and CSH-Px decreased,MDA level increased,Bax expression increased and Bcl-2 decreased in the APAP group,and the H&E staining results showed significant liver damage.Compared with APAP group,ALT,AST,MDA and Bax levels in L-BA and H-BA groups were decreased,while CSH,Gsh-PX and Bcl-2 levels were increased.H&E staining showed that liver injury was significantly alleviated.In conclusion,BA had a protective effect on APAP-induced acute liver injury in mice,and its mechanism might be related to alleviating the imbalance of free radical-antioxidant oxidase caused by oxidative stress in mice liver and regulating the expression of apoptosis-related proteins Bax and Bcl-2.
