摘要
目的探讨胃癌中血小板衍生生长因子受体α(platelet-derived growth factor receptor alpha,PDGFRB)对胃癌患者预后的影响,并分析它和肿瘤免疫的相关性,为胃癌免疫治疗提供新思路。方法在公共数据库中下载胃癌患者的基因表达谱数据集、突变数据和相关临床信息进行后续分析。本研究首先比较了PDGFRB在不同肿瘤中的表达和预后情况。同时,还探究了PDGFRB和免疫检查点、肿瘤突变负荷(tumor mutational load,TMB)以及免疫微环境之间的关系,并用这些特征构建了列线图。用基因集富集分析(GSEA)探索PDGFRB高低表达组显著富集的通路,并用TIDE算法评价PDGFRB高低表达组对免疫治疗的反应。结果PDGFRB在胃癌患者中高表达,并且可以作为胃癌的独立预后危险因素。PDGFRB高表达组具有较低的TMB,较高的免疫评分和基质评分,以及较高的免疫检查点表达量。产生IgA的肠道免疫网络等信号通路在PDGFRB高表达组显著富集。PDGFRB高表达组可能对免疫治疗的反应更好。结论PDGFRB可作为潜在的分子标志物来预测胃癌的预后情况并为胃癌的免疫治疗提供新思路。
Objective To investigate the effect of platelet-derived growth factor receptor alpha(PDGFRB)in gastric cancer on the prognosis of gastric cancer patients and to analyze its correlation with tumor immunity to provide new ideas for gastric cancer immunotherapy.Methods Gene expression profile datasets,mutation data and relevant clinical information of gastric cancer patients were downloaded from public database for subsequent analysis.Firstly compared the expression and prognosis of PDGFRB in different tumors.Also,explored the relationship between PDGFRB and immune checkpoints,tumor mutational load(TMB)and immune microenvironment.And these features were used to construct a nomogram.Exploring the pathways significantly enriched in PDGFRB high and low expression groups using GSEA enrichment analysis.Results PDGFRB was highly expressed in gastric cancer patients and could be an independent prognostic risk factor for gastric cancer.The PDGFRB high expression group had a lower TMB,higher immune score and stromal score,and higher immune checkpoint expression.Signaling pathways such as the intestinal immune network for IgA production were significantly enriched in the PDGFRB high expression group.PDGFRB high expression group might respond better to immunotherapy.Conclution PDGFRB could be a potential molecular marker to predict the prognosis of gastric cancer and provide new ideas for immunotherapy of gastric cancer.
