miR-10b-5p在腹主动脉瘤患者血清的表达及临床意义

Expression and clinical significance of serum miR-10b-5p in patients with abdominal aortic aneurysm

目的探讨微小RNA-10b-5p(miR-10b-5p)在腹主动脉瘤(AAA)患者血清中的表达及临床意义。方法回顾性分析40例AAA患者(AAA组)和40例健康体检者(对照组)的临床资料,采用定量RT-PCR检测血清miR-10b-5p、IL-6和IL-10表达,分析miR-10b-5p表达与相关临床指标的相关性,并评估miR-10b-5p表达对AAA发生的诊断价值。结果与对照组相比,AAA组淋巴细胞计数、IL-10表达、TC和HDL-C水平降低,中性粒细胞计数、白细胞计数、miR-10b-5p和IL-6表达以及TG、LDL-C、脂蛋白a、D-D、CRP水平升高(P<0.05)。AAA组miR-10b-5p表达与LDL-C水平和IL-6表达呈正相关(r=0.271、0.642,P<0.05),而与HDL-C水平和IL-10表达呈负相关(r=-0.549、-0.456,P<0.05)。miR-10b-5p诊断AAA发生的AUC为0.909[95%CI(0.841~0.976),P<0.05],当取最佳诊断界值为1.007时,其对应的诊断灵敏度和特异度分别为80%和95%。结论miR-10b-5p在AAA患者血清中表达增高,与部分血脂及炎症因子指标相关,对AAA具有较高的诊断效能。

Objective To investigate the expression and clinical significance of serum miR-10b-5p in the patients with abdominal aortic aneurysm(AAA).Methods The clinical data of 40 patients with AAA(group A)and 40 healthy people(group C)were retrospectively analyzed.The expressions of serum miR-10b-5p,IL-6 and IL-10 were detected by quantitative RT-PCR.The correlation between miR-10b-5p expression and related clinical indexes was analyzed,and the diagnostic value of miR-10b-5p in the occurrence of AAA was evaluated.Results Compared with group C,the lymphocyte count,IL-10 expression and TC and HDL-C levels were decreased,while the neutrophil count,WBC count,expressions of miR-10b-5p and IL-6 and the levels of TG,LDL-C,lipoprotein(a),D-D and CRP were increased in group A(P<0.05).The miR-10b-5p expression was positively correlated with LDL-C level and IL-6 expression(r=0.271 and 0.642,P<0.05),and negatively correlated with HDL-C level and IL-10 expression in group A(r=-0.549 and-0.456,P<0.05).The AUC value of miR-10b-5p in the diagnosis of AAA was 0.909[95%CI(0.841-0.976),P<0.05],and taking 1.007 as the optimal cut-off value,the corresponding diagnosis sensitivity and specificity were 80%and 95%,respectively.Conclusion The expression of miR-10b-5p is increased in the serum of AAA patients,which is associated with some blood lipids and inflammatory factors.miR-10b-5p has a higher diagnostic efficiency for AAA.
