解淀粉芽孢杆菌NTBA分离及对小白菜的促生活性测定

摘要 为挖掘优良的促生细菌菌株,通过形态学和分子生物学方法对烟草中分离的内生细菌进行鉴定,并测定其对小白菜的促生活性。经鉴定该内生菌为解淀粉芽孢杆菌(Bacillus amyloliquefaciens),命名菌株为NTBA。该菌具有解有机磷、解无机磷和解钾活性,并能够产IAA。采用土壤中施入100、10、1μL菌液的浇灌施菌法和小白菜种子的菌液浸种法,并以浇灌无菌水作为对照,测定对小白菜的促生作用。结果表明,培养10、15和20 d,2种施菌方法处理的小白菜的叶长、叶宽、株高及茎粗均显著高于对照;培养20 d时,3个浓度菌液浇灌处理和菌液浸种处理的小白菜鲜质量分别比对照增加了78.99%、76.08%、70.29%和84.78%,干质量增加了86.44%、76.27%、31.36%和88.98%,最长根长分别增加了118.98%、83.94%、45.99%和63.50%。可见,解淀粉芽孢杆菌NTBA能够明显促进小白菜生长,提高其产量。研究结果可为小白菜生产提供有益的促生菌,也为其他作物生产提供了潜在的促生菌资源。 To explore excellent strains of growth promoting bacteria,endophytic bacteria isolated from tobacco were identified using morphological and molecular biology methods,and its growth promoting activity on Chinese cabbage was determined.The endophytic bacterium was identified as Bacillus amyloliquefaciens,and assigned as NTBA.It was found that the bacterium had the activity of hydrolyzing organic phosphorus,inorganic phosphorus and potassium,and could produce IAA.The growth promotion effect on Chinese cabbage was determined by pouring 100μL,10μL and 1μL bacterial solutions in the soil and by soaking Chinese cabbage seeds with bacterial solution,with sterile water as the control.The results showed that after cultured for 10,15,and 20 days,the leaf length,leaf width,plant height,and stem diameter of Chinese cabbage treated with the two methods were significantly higher than those of the control.After 20 days of culture,compared with the control,the fresh mass of Chinese cabbage treated by pouring three concentrations of the bacterial solution and soaking seeds with bacterial solution increased by 78.99%,76.08%,70.29%,and 84.78%,respectively.The dry mass increased by 86.44%,76.27%,31.36%,and 88.98%,respectively.The root growth of Chinese cabbage was increased by 118.98%,83.94%,45.99%,and 63.50%,respectively.In conclusion,B.amylolyquenfaciens NTBA can obviously promote the growth of Chinese cabbage and increase its yield.This study provides beneficial plant growth-promoting bacteria for the production of Chinese cabbage and potential growth-promoting bacteria resources for other crops.
