合成耐盐菌群对水稻品种五山丝苗耐盐性的影响

Effects of Synthetic Salt-tolerant Bacteria on Salt Tolerance of Rice Variety Wushansimiao

摘要 为了提高水稻的耐盐能力,构建了由5个新筛选的耐盐细菌菌株(L-A8、L-B2、L-B9、L-D6和L-D7)和1个现有铜绿假单胞菌(Pseudomonas aeruginosa)菌株(L3)组成的人工合成菌群(Artificially synthetic community,ASC),探究其对水稻品种五山丝苗耐盐性的影响。结果表明,通过16S rRNA序列鉴定,L-A8为无色杆菌属(Achromobacter),L-B2和L-B9为肠杆菌属(Enterobacter),L-D6和L-D7为志贺氏菌属(Shigella)。与对照组相比,ASC处理后的种子发芽势和成苗率分别显著提高41.09和21.78个百分点,根长和芽长分别显著增加161.9%、201.7%。在水培条件下,与对照组相比,ASC处理的幼苗鲜重、叶绿素a和叶绿素b的含量分别显著增加107.8%、127.9%和118.1%,幼苗根部的超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)含量分别显著提高75.9%、41.4%和35.4%,丙二醛(MDA)含量显著降低37.0%。在盆栽条件下,与对照组相比,ASC处理的幼苗根长、鲜重、叶绿素a以及叶绿素b含量分别显著增加84.1%、65.0%、48.6%和65.9%,SOD、POD和CAT活性分别显著提高16.8%、78.7%和23.0%,MDA含量显著降低23.2%。与对照组相比,ASC处理的五山丝苗在盐胁迫下的有效穗数增加8.8%,每穗总粒数和每穗实粒数分别显著增加22.5%和39.1%,结实率显著提高9.68个百分点,最终使得单株产量显著提高63.1%。综上,ASC对提高盐碱地的利用率具有现实意义,可作为一种有效提升盐碱地应用价值的手段。 To improve the salt tolerance of rice,an artificially synthetic community(AsC)was constructed,consisting of five newly selected salt-tolerant bacterial strains(L-A8,L-B2,L-B9,L-D6 and L-D7)and one existing strain of Pseudomonas aeruginosa(L3),to investigate its effects on salt tolerance of rice variety Wushansimiao.The results showed that L-A8 was identified by 16S rRNA sequence as Achromobacter,L-B2 and L-B9 as Enterobacter,and L-D6 and L-D7 as Shigella.Compared with the control group,the seed germination potential and adult seedling rate after ASC treatment were significantly increased by 41.09 and 21.78 percentage points,and the root length and bud length were significantly increased by 161.9% and 201.7%,respectively.Under hydroponic conditions,compared with the control group,the fresh weight,content of chlorophyll a and content of chlorophyll b of ASC treated seedlings were significantly increased by 107.8%,127.9% and 118.1%,respectively.The contents of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT)were significantly increased by 75.9%,41.4% and 35.4%,respectively,while the content of malondialdehyde(MDA)was significantly decreased by 37.0% in the roots of seedlings.Under pot conditions,compared with the control group,the root length,fresh weight,content of chlorophyll a and content of chlorophyll b of ASC treated seedlings were significantly increased by 84.1%,65.0%,48.6% and 65.9%,respectively,and the activities of SOD,POD and CAT were significantly increased by 16.8%,78.7% and 23.0%,respectively,and MDA content was significantly decreased by 23.2%.Compared with the control group,the number of effective panicles,number of total spikelets per panicle and number of filled grains per panicle were increased by 8.8%,22.5% and 39.1%,respectively,and the seed setting rate was increased by 9.68 percentage points,resulting in a significantly increased yield per plant by 63.1%.In summary,ASC has practical significance to improve the utilization rate of saline-alkali land,and can be used as a means to effectively enhance the application value of saline-alkali land.
