摘要
目的探究甲状腺癌与其他癌症之间的相互关系,为了及早发现甲状腺癌患者在监测随访过程中第二原发性癌的发生,对与具有高风险继发于甲状腺癌的第二原发性癌进行诊断性筛查提供有力依据,进行早期诊断,增加甲状腺癌患者生存率。方法本研究采用公开获得来源于欧洲的GWAS数据进行孟德尔随机化分析,使用Cochran Q检验异质性,采用MR-Egger回归、加权中位数法(weight median estimator,WMR)、随机效应逆方差加权法(inverse-variance weighted,IVW)三种回归模型对甲状腺癌与其他癌症之间的相互关系进行孟德尔随机化分析。结果利用纳入989例甲状腺癌的GWAS数据和8个癌症数据库进行孟德尔随机化分析,结果发现甲状腺癌与乳腺癌(OR=1.02,95%CI=1.00~1.03,P=0.016)、前列腺癌(OR=1.02,95%CI=1.00375~1.037171,P=0.016)、非霍奇金淋巴瘤(OR=1.1397,95%=1.003518~1.294412,P=0.04)之间存在弱相关性。结论甲状腺癌与乳腺癌、前列腺癌及非霍奇金淋巴瘤可能相关,未发现与其他癌症具有相关性。
Objective To explore the correlation between thyroid cancer and secondary cancers,in order to early detect the second primary cancers in thyroid cancer patients during monitoring and follow-up,and provide strong evidence for diagnostic screening of the second primary cancers those with a high risk of being secondary to thyroid cancer,to realize early diagnosis and increase the survival rate of thyroid cancer patients.Methods This study adopted publicly available GWAS data from Europe for Mendelian randomization analysis,conducted Cochran Q test for heterogeneity,and three regression models-MR-Egger regression,weighted median estimator(WME),and inverse-variance weighted(IVW)regression were employed to analyze the correlation between thyroid cancer and secondary cancers.Results The Mendelian randomization analysis using GWAS data from 989 cases of thyroid cancer and 8 cancer databases revealed a weak correlation between thyroid cancer and breast cancer(OR=1.02,95%CI=1.00~1.03,P=0.016),prostate cancer(OR=1.02,95%CI=1.00375-1.037171,P=0.016),and non-Hodgkin's lymphoma(OR=1.1397,95%CI=1.003518~1.294412,P=0.04).Conclusions Thyroid cancer may be correlated with breast cancer,prostate cancer,and non-Hodgkin's lymphoma,with no correlation to other cancers observed.
