摘要
目的探讨外周血单个核细胞(PBMC)中泛素折叠修饰因子1(UFM1)、生长分化因子15(GDF15)的表达水平与冠心病之间的关系以及UFM1与GDF15之间表达的相关性。方法收集2020年12月至2021年5月在北京大学人民医院心血管内科就诊且经冠状动脉造影检查的患者181例,其中排除冠心病的患者16例(非冠心病组)、确诊冠心病患者165例(冠心病组)。收集研究对象外周血并提取PBMC中总RNA,分析PBMC中GDF15和UFM1的表达差异。结果GDF15在非冠心病组和冠心病组相对表达水平分别为0.694(1.667)、1.275(2.082),冠心病组GDF15表达水平显著升高(P=0.0423)。UFM1在非冠心病组和冠心病组相对表达水平分别为1.088(1.618)、1.675(1.206),冠心病组UFM1表达水平显著升高(P=0.0034)。UFM1与GDF15表达水平呈正相关(r=0.4435,P<0.0001)。多因素logistic回归显示,年龄、高脂血症以及UFM1表达水平是冠心病的危险因素,UFM1表达水平升高增加冠心病患病风险(OR=4.170,P=0.009)。结论PBMC中UFM1水平升高与冠心病关系密切,可能对冠心病的临床诊疗有一定指导价值。
Objective To investigate the expression of growth differentiation factor 15(GDF15)and ubiquitin-fold modifier 1(UFM1)in peripheral blood mononuclear cells(PBMCs)in patients with coronary heart disease(CHD)and the correlation between the expressions of them.Methods A total of 181 patients who visited the Department of Cardiology at our hospital from December 2020 to May 2021 and underwent coronary angiography were collected.Among them,16 patients with coronary heart disease were excluded(NC group)and 165 patients with confirmed coronary heart disease(CHD group).Peripheral blood was collected and total RNA was extracted from PBMC,and the expression level of GDF15 and UFM1 in PBMC was compared.Results The relative expression levels of GDF15 in the NC and CHD groups were 0.694(1.667)and 1.275(2.082),while the relative expression of UFM1 in NC and CHD groups were 1.088(1.618)and 1.675(1.206),respectively.Compared with the NC group,the expression levels of GDF15 and UFM1 in the CHD group were both significantly increased(P=0.0423,P=0.0034).The expression of UFM1 was positively correlated with GDF15 in PBMCs(r=0.4435,P<0.0001).Multivariate logistic regression showed that age,hyperlipidemia,and UFM1 expression levels were risk factors for CHD.High UFM1 level increased the risk of CHD(OR=4.170,P=0.009).Conclusion The increased level of UFM1 in PBMC is closely related to CHD,and may have certain guiding value for the clinical diagnosis and treatment of CHD.
