不同部位血管舒缩反应性与多不饱和脂肪酸的血管活性作用分析

Vascular Contraction and Relaxation Responsiveness of Different Vascular Beds and Effects of Polyunsaturated Fatty Acids

摘要 目的评价小鼠不同部位血管床的收缩和舒张反应性,比较多不饱和脂肪酸花生四烯酸(AA)、二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)的血管活性作用。方法分离小鼠股动脉(FA)、主动脉(AO)、肠系膜动脉(MA)、冠状动脉(CA),用微血管张力仪评价其对苯肾上腺素(PE)、U46619的收缩反应性和对乙酰胆碱(ACh)的舒张反应性,拟合剂量-效应曲线,对内皮功能良好的血管进行评价,比较AA、EPA、DHA的作用。结果各血管对U46619的收缩程度比对PE的收缩程度大,PE不收缩CA。以U46619作预收缩剂,比较各血管对ACh的舒张反应,发现MA的舒张程度最大,CA的舒张最不敏感。3种多不饱和脂肪酸诱导CA舒张的程度最小,诱导MA舒张的程度最大、最敏感。与AA和EPA相比,DHA诱导各血管的舒张作用更敏感。结论小鼠不同部位血管床的舒缩反应性有明显差异,各血管均显示对3种多不饱和脂肪酸的舒张反应,其中对DHA的舒张效应最敏感。 Objective To evaluate contraction and relaxation responses of different vascular beds of mice,and to compare the vasoactive effects of the polyunsaturated fatty acids-arachidonic acid(AA),eicosapentaenoic acid(EPA),and docosahexaenoic acid(DHA).Methods Femoral artery(FA),aorta(AO),mesenteric artery(MA),and coronary artery(CA)of male and female mice were isolated.Vasocontraction to phenylephrine(PE)and U46619 and vasorelaxation responses to acetylcholine(ACh)were measured by wire myography,with each dose-effect curve fitted,and responses to AA,EPA,or DHA were evaluated and compared in vessels with good endothelial function.Results Each vessel contracted greater to U46619 than to PE,which did not contract CA.Comparing the vasorelaxation responses to ACh among vessels,MA exhibited the greatest vasorelaxation and CA exhibited the least potency.CA exhibited the least vasorelaxation,and MA exhibited the greatest vasorelaxation and the least potency.Compared with AA and EPA,DHA exhibited less potency in each vessel.Conclusion There is significant difference in vasocontraction and vasorelaxation to different vascular beds of mice.Three polyunsaturated fatty acids induce vasorelaxation to all vessels,and DHA exhibitis the greatest potency of vasorelaxation.
