摘要
目的探讨长链非编码RNA(lncRNA)SNHG14的表达在急性冠状动脉综合征(ACS)患者中的价值和潜在机制。方法选取2019年11月至2021年8月该院收治的84例ACS患者为研究对象,包括28例STEMI患者、31例NSTEMI患者和25例UA患者,选取同期经冠状动脉造影证实冠状动脉正常的75例患者作为对照组。使用实时荧光定量PCR技术检测SNHG14表达水平,绘制受试者工作特征(ROC)曲线评价SNHG14的诊断价值。所有患者随访6个月,对影响预后不良的因素进行Cox回归分析,评估SNHG14的预后价值。通过CCK-8、流式细胞术和ELISA检测SNHG14对HUVEC细胞模型的调控作用,荧光素酶报告实验用于验证SNHG14的靶向基因。结果ACS患者的SNHG14表达水平增加。低表达水平SNHG14可以缓解H/R对细胞活力、凋亡和炎症的不良影响。miR-30c-5p是SNHG14的1个靶基因,在ACS患者中表达水平降低。在ACS患者中,SNHG14和miR-30c-5p的表达水平成负相关。miR-30c-5p介导SNHG14对HUVEC细胞模型的调控。结论SNHG14表达水平对预测ACS的发生和短期预后有很高的价值,SNHG14/miR-30c-5p轴可以通过调控HUVEC细胞的生物功能。
Objective To investigate and analyze the value of lncRNA SNHG14 expression in patients with acute coronary syndrome(ACS).Methods 84 patients with ACS admitted to this hospital from November 2019 to August 2021 were selected as the study objects,including 28 patients with STEMI,31 patients with NSTEMI and 25 patients with UA,and 75 patients with normal coronary arteries confirmed by coronary angiography during the same period were selected as the control group.The relative levels of SNHG14 were detected by qRT-PCR.Receiver operating characteristic(ROC)curve was drawn to evaluate the possibility of SNHG14 in predicting the onset of ACS.Then,all patients were followed up for 6 months,and Cox regression analysis was conducted to evaluate the short-term prognostic value of SNHG14.CCK-8,flow cytometry,and ELISA were used to examine the regulatory effect of SNHG14 on HUVEC cell model.Luciferase reporter assay was used to verify the target genes of SNHG14.Results The expression of SNHG14 was increased in ACS patients.Low expression levels of SNHG14 could alleviate the adverse effects of H/R on cell viability,apoptosis and inflammation by targeting miR-30c-5p.The level of miR-30c-5p was reduced in ACS patients and negatively correlated with the level of SNHG14.Conclusion SNHG14 expression level was a great value in predicting the occurrence and short-term prognosis of ACS patients.SNHG14/miR-30c-5p axis might be involved in the development of ACS by regulating the biological function of HUVEC cells.
