解毒复正合剂的提取工艺优化及指纹图谱研究

摘要 目的优化解毒复正合剂的水提工艺,建立质量标准及指纹图谱对其化学成分进行鉴定。方法采用正交试验方法,L_(9)(3^(4))试验设计考察加水量、提取次数、提取时间等因素对提取工艺的影响,优选解毒复正合剂的水提工艺;采用HPLC法建立其含量测定方法,同时建立其指纹图谱,以黄芩苷、橙皮苷总含量,出膏率及总黄酮含量为综合评价指标。结果确定解毒复正合剂最佳水提工艺是以10倍水,煎煮3次,每次1.5h;紫外分光光度法结果显示,解毒复正合剂总黄酮平均含量为33.423μg/ml,HPLC结果显示,黄芩苷、橙皮苷平均总含量为771.971μg/ml。结论该研究为解毒复正合剂水提工艺的确定提供了依据,建立的质量标准也能稳定的控制其有效物质成分,为后续研究打下了基础。 Objective To optimize the water extraction process of detoxification and rejuvenating mixture,and establish quality standards and fingerprints to identify its chemical composition.Methods The orthogonal test method and L9(34)experimental design were used to investigate the effects of water addition,extraction times,extraction time and other factors on the extraction process,and the water extraction process of the detoxification and renormalization mixture was optimized.The content determination method was established by HPLC method,and the fingerprint was established,and the total content of baicalin,hesperidin,ointment yield and total flavonoids were used as comprehensive evaluation indexes.Results The best water extraction process was determined to be 10 times of water,decoction for 3 times,each time for 1.5h,ultraviolet spectrophotometry results showed that the average content of total flavonoids of detoxification and rejuvenating mixture was 33.423μg/ml,and HPLC results showed that the average total content of baicalin and hesperidin was 771.971μg/ml.Conclusion This study provides a basis for the determination of the water extraction process of detoxification and rejuvenating mixture,and the established quality standard can also stably control its active substance composition,which lays a foundation for subsequent research.
