摘要
采用超声辅助天然低共熔溶剂(NDES)法提取柚皮总黄酮。在筛选出最佳NDES的基础上,通过单因素试验和正交试验进一步优化总黄酮提取工艺,并对其抗氧化活性进行评价。结果显示:以氯化胆碱-甘油-乳酸为提取溶剂,在料液比1∶35(g/mL)、加水量30%(以NDES体积计)、超声时间15 min、超声温度60℃的条件下,得到总黄酮提取量最高为17.999 mg/g。在一定质量浓度范围内,提取的柚皮总黄酮对DPPH自由基清除率、亚铁离子螯合率、羟基自由基清除率、铁离子还原力和脂质过氧化抑制率最高分别为92.36%、42.93%、87.94%、0.513和22.43%,表明柚皮总黄酮具有较好的抗氧化活性。
Total flavonoids from pomelo peel were extracted by ultrasonic-assisted natural deep eutectic solvent(NDES).On the basis of selecting the best NDES,the extraction process of total flavonoids was further optimized by single factor experiments and orthogonal experiment,and its antioxidant activity was evaluated.The results showed that the maximum total flavonoids extraction was 17.999 mg/g under the conditions of material-liquid ratio 1:35(g/mL),water content 30%(by volume of NDES),ultrasound time 15min,and ultrasonication temperature 60℃,using choline chloride-glycerol-lactic acid as the extraction solvent.Within a certain mass concentration range,the highest DPPH radical scavenging,chelation rate of ferous ions,scavenging rate of hydroxyl radical,reducing power,and inhibition rate of lipid peroxidation of total flavonoids from pomelo peel were 92.36%,42.93%,87.94%,0.513 and 22.43%,respectively,indicating that total flavonoids from pomelo peel had good antioxidant activity.
