草莓叶斑病真菌性病原菌的分离与鉴定

摘要 从新疆和静县巴润哈尔莫墩镇草莓棚采集具有代表性的病叶,采用组织分离法从病株将病菌分离出来,在PDA上分离纯化获得纯种菌株,先利用致病性实验鉴定病原菌,再利用试剂盒提取DNA,通过引物(ITS1和ITS4)进行PCR扩增。得到序列后,将序列复制到NCBI官网中,进行Blast分析,在GenBank中进行同源性比对,下载同源性比较近的菌株,再利用MAGE7.0构建系统发育树。经过鉴定得知,采样所得的2种叶斑病的病原菌是木贼镰刀菌,为防治草莓病害提供数据支持。 Representative diseased leaves were collected from greenhouse,and the pathogens were isolated from the diseased plants by tissue separation method.The pure strains were isolated and purified on PDA,and the pathogenic bacteria were identified by pathogenicity test.DNA was extracted using kits and amplified by PCR using primers(ITS1 and ITS4).After obtaining the sequences,the sequences were copied to the NCBI official website for Blast analysis,homology comparison was performed in GenBank,and the strains with close homology were downloaded.Phylogenetic tree was constructed by MAGE 7.0.After identification,it was found that the pathogen of two kinds of anthracnose was Fusarium equiseti,to provide data support for the prevention and control of strawberry diseases.
