不同白油佐剂制备的鸡新城疫、禽流感(H9亚型)二联灭活疫苗的比较试验

摘要 为研究不同白油佐剂制备的灭活疫苗免疫效果,试验采用2个厂家的3个国外进口白油样品、1个国产白油样品,制备4批鸡新城疫、禽流感(H9亚型)(La Sota株+WD株)二联灭活疫苗(以下简称:鸡新-流二联灭活疫苗),分别标A、B、C、D,按照现行版《中华人民共和国兽药典》方法,进行性状、安全检验、抗体检测以及吸收情况对比。结果表明,4批样品析出量均小于0.1 mL,稳定性合格。黏度对比结果,进口白油B最低27.6 cP、进口A 48.2 cP、进口C 56.5cP、国产D 52.0 cP,吸收情况D组最好,其次是C、A,B组最差。免疫持续期对比结果B组抗体最好,免疫后28 d抗体最高,ND抗体滴度为9.83log2,H9抗体滴度为11.33log2,直到免疫后154 d,ND抗体滴度为5.1log2,H9抗体滴度为8.6log2,仍可以达到保护效果;其次是D组,ND部分免疫后21 d抗体滴度最高为8.17log2,H9免疫后42 d抗体滴度最高为9.58log2,ND免疫后154 d抗体滴度为4.2log2,H9抗体滴度为7.8log2;最后为C、A两组。综合评定B组抗体滴度最高但吸收度最差,蛋鸡免疫时可以考虑使用;D组抗体和吸收整体比较好,结合性价比确定为最佳白油佐剂。 In order to study the immune effect of inactivated vaccines prepared with different white oil adjuvants,three foreign white oil samples and one domestic white oil sample from two manufacturers were used to prepare four batches of chicken Newcastle disease and avian influenza(H9 subtype)(La Sota+WD)bivalent inactivated vaccine(hereinafter referred to as:Newcastle disease and avian influenza bivalent inactivated vaccine)[1],Marks A,B,C,D,respectively according to the current edition of the'Veterinary Pharmacopoeia of the People's Republic of China'method[2],the characteristics,safety test,antibody detection and absorption comparison were carried out.The antibody was detected by subcutaneous injection of 0.3 ml in the neck.The results showed that the precipitation amount of 4 batches of samples was less than 0.1ml,and the stability was qualified.The lowest viscosity of imported white oil B was 27.6cP,imported A:48.2cP,imported C:56.5cP,domestic D:52.0cP,and the absorption of group D was the best,followed by group C,A and B,and group B was the worst.Compared with the duration of immunization,the antibody in group B was the best.The highest ND antibody was 9.83 log2 and H9 was 11.33 log2 on the 28 th day after immunization.Until 154 days after immunization,the ND antibody was 5.1 log2 and H9 was 8.6 log2,which could still achieve the protective effect.The second was group D.The highest ND antibody was 8.17 log2 on the 21 st day after immunization,and the highest H9 antibody was 9.58 log2 on the 42 nd day after immunization.Until 154 days after immunization,ND antibody was 4.2 log2,H9 was 7.8 log2,and finally C and A groups.Comprehensive evaluation Although the antibody of group B was the highest but the absorption was the worst,the immune use of laying hens can be considered.The antibody and absorption of group D were better as a whole,and then combined with the cost performance,it was determined as the best white oil adjuvant.
