山西杓兰(Cypripedium shanxiense)内生真菌种类的鉴定

Identification of Endophytic Fungi Species from Cypripedium shanxiense

摘要 本研究对山西分布的山西杓兰内生真菌种类进行分析,并与已报道的其他地区分布的山西杓兰内生真菌种类进行比较,探讨地理分布与山西杓兰内生真菌种类相关性。通过对内生真菌ITS基因片段测序并从GenBank下载相似序列,构建NJ系统发育树,根据相似性原则,对内生真菌进行分子鉴定。结果表明,山西杓兰内生真菌包括11个代表性分类单元,分属4纲:伞菌纲(Agaricomycetes)(36%),座囊菌纲(Dothideomycetes)(28%),粪壳菌纲(Sordariomycetes)(18%),散囊菌纲(Eurotiomycetes)(18%);5目:多孔菌目(Polyporales)(28%),伞菌目(Agaricales)(9%),格孢腔菌目(Pleosporales)(18%),肉座菌目(Hypocreales)(9%),散囊菌目(Eurotiales)(18%),未知目(18%)。山西杓兰内生真菌与其他地区的山西杓兰内生真菌种类有一定差异性,说明地理分布与内生真菌种类组成具有一定的相关性。 This study analyzed the species of endophytic fungi in Cypripedium shanxiense in Shanxi Province, and compared with earlier reports about C. shanxiense from other regions distribution to explore the correlation between geographic distribution and the species of endophytic fungi in C. shanxiense. Through sequencing the ITS gene fragments of endophytic fungi and downloading similar sequences from GenBank, an NJ phylogenetic tree was constructed. Based on the principle of similarity, the endophytic fungi were identified molecularly. The results showed that the endophytic fungi in Cypripedium shanxiense include 11 representative taxa belonging to 4 classes:Agaricomycetes(36%), Dothideomycetes(28%), Sordariomycetes(18%), Eurotiomycetes(18%);5 orders: Polyporales(28%), Agaricales(9%), Pleosporales(18%), Hypocreales(9%), Eurotiales(18%), not assigned order(18%). There is a certain difference in the species of endophytic fungi in C. shanxiense compared with other regions,indicating that geographic distribution has a certain correlation with the composition of endophytic fungal species.
