高效液相色谱分析大高良姜、红豆蔻乙酸乙酯部位血清药物化学研究

摘要 目的应用高效液相色谱(High performance liquid chromatography,HPLC)分析空白模型大鼠及胃溃疡寒证模型大鼠的血清药物成分,探讨大高良姜、红豆蔻乙酸乙酯部位抗胃溃疡寒证作用的药效物质基础。方法采用乙醇回流提取法分别提取并纯化富集得到大高良姜和红豆蔻乙酸乙酯部位浸膏;用灌服冰乙酸及4℃冰知母水煎液法复制胃溃疡寒证模型;通过HPLC法对大高良姜、红豆蔻乙酸乙酯部位进行基于空白模型大鼠及胃溃疡寒证模型大鼠的血清药物化学研究。结果优化确定大高良姜、红豆蔻含药血清处理方法,同时标识出10个空白模型大鼠含药血清共同入血成分,其中7个为原型入血成分;标识出12个胃溃疡寒证模型大鼠含药血清入血成分,其中8个为共同入血成分。结论大高良姜、红豆蔻乙酸乙酯部位均具有相似的抗胃溃疡寒证作用机制,同时,初步推测出原儿茶酸、原儿茶醛、对羟基苯甲酸、芦丁、对羟基苯甲醛、对羟基桂皮酸可能为大高良姜、红豆蔻乙酸乙酯部位抗胃溃疡寒证作用的物质基础。 Objective High performance liquid chromatography(HPLC)was used to analyze the serum drug components of blank model rats and gastric ulcer cold syndrome model rats and explore the pharmacodynamic material basis of the effect of treating gastric ulcer cold syndrome effect of the ethyl acetate extracts of Dagaoliangjiang(Alpinia galangal)and Hongdoukou(Alpinia galanga Willd.).Methods The ethanol reflux extraction method was used to extract and purify the ethyl acetate extracts of Dagaoliangjiang(Alpinia galangal)and Hongdoukou(Alpinia galanga Willd.).The gastric ulcer cold syndrome model was replicated by intragastric administration of glacial acetic acid and 4℃ice anemarrhenae decoction.The serum pharmacochemistry of the ethyl acetate extracts of Dagaoliangjiang(Alpinia galangal)and Hongdoukou(Alpinia galanga Willd.)based on blank model rats and gastric ulcer cold syndrome model rats was studied by HPLC.Results The treatment method of drug-containing serum of Dagaoliangjiang(Alpinia galangal)and Hongdoukou(Alpinia galanga Willd.)was optimized and determined.At the same time,10 common blood components of drug-containing serum in blank model rats were identified,of which 7 were prototype blood components.A total of 12 blood components of gastric ulcer cold syndrome model rats were identified,of which 8 were common blood components.Conclusion The ethyl acetate extracts of Dagaoliangjiang(Alpinia galangal)and Hongdoukou(Alpinia galanga Willd.)have similar anti-gastric ulcer cold syndrome mechanism.It is preliminarily inferred that protocatechuic acid,protocatechuic aldehyde,p-hydroxybenzoic acid,rutin,p-hydroxybenzaldehyde and p-hydroxycinnamic acid may be the material basis for the anti-gastric ulcer syndrome of the ethyl acetate extracts of Dagaoliangjiang(Alpinia galangal)and Hongdoukou(Alpinia galanga Willd.).
