山茱萸炭有效成分的含量测定及止血作用研究

摘要 目的分析山茱萸炭与山茱萸中有效成分的含量变化及山茱萸炭对止血作用的影响,为阐明山茱萸炭作用机制奠定基础。方法通过采用高效液相测定生品和炒炭后山茱萸的马钱苷、莫诺苷、熊果酸含量变化,采用苯酚-硫酸法和干酪素-磷钼钨酸法测定山茱萸多糖和没食子酸的含量,测定不同剂量的山茱萸炭对小鼠凝血时间、活化部分凝血活酶时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)和纤维蛋白原含量(FIB)。结果山茱萸炭中莫诺苷含量显著减少,马钱苷、熊果酸含量略有增加,没食子酸含量显著增加,山茱萸多糖含量基本不变。另外,中、低剂量组的山茱萸炭可以显著降低小鼠的凝血时间,可以显著降低小鼠的TT。结论山茱萸炭可以在一定程度上促进小鼠的凝血,表现出一定炒炭止血的特点。本研究有效拓展了山茱萸的炮制方法和功效范围、发展了中药炭药的种类,具有重要的应用和理论价值。 Objective To analyze the changes in the content of active ingredients in Cornus officinalis charcoal and its effect on hemostasis,laying the foundation for elucidating the mechanism of action of Cornus officinalis charcoal.Method The content changes of loganin,mononucleoside,and ursolic acid in Shanzhuyu(Corni Fructus)charcoal and Shanzhuyu(Corni Fructus)were determined by high-performance liquid chromatography.The content of polysaccharides and gallic acid in Shanzhuyu(Corni Fructus)charcoalwas determined by phenol sulfuric acid method and casein phosphomolybdic acid method.The coagulation time,activated partial thromboplastin time(APTT),prothrombin time(PT),thrombin time(TT),and fibrinogen content(FIB)were measured in mice.Result The content of mononucleoside in Shanzhuyu(Corni Fructus)charcoal significantly decreased,while the content of loganin and ursolic acid slightly increased,and the content of gallic acid significantly increased.The content of polysaccharides in Shanzhuyu(Corni Fructus)charcoal remained basically unchanged.In addition,the middle and low dose groups of Shanzhuyu(Corni Fructus)charcoalcan significantly reduce the coagulationtime and TT of mice.Conclusion Shanzhuyu(Corni Fructus)charcoal can promote coagulation in mice to a certain extent,exhibiting certain characteristics of charcoal roasting for hemostasis.This study effectively expanded the processing methods and efficacy range of Shanzhuyu(Corni Fructus)charcoal,developed the types of traditional Chinese charcoalmedicine,and has important application and theoretical value.
