表儿茶素没食子酸酯对免疫低下小鼠免疫功能的调节作用

Modulatory effect of epigallocatechin gallate on immune function in immunocompromised mice

摘要 研究表儿茶素没食子酸酯(EGCG)对免疫低下小鼠免疫功能的影响。利用环磷酰胺构建免疫低下小鼠模型,测定EGCG对免疫低下小鼠免疫器官指数及脾脏组织病理变化,检测免疫细胞核转录因子PAX5、GATA3表达水平及NFκB、AKT、MAPK信号通路关键分子蛋白表达水平。结果表明,EGCG干预后的小鼠脾脏指数(1.95 g/kg)、胸腺指数(0.73 g/kg)与模型组(1.43,0.47 g/kg)相比均显著上升(P<0.05),EGCG可改善免疫低下小鼠的脾脏质量/指数;切片结果显示,EGCG能够缓解免疫低下小鼠脾脏的损伤;部分恢复了B细胞中PAX5和T细胞中GATA3 mRNA的表达(P<0.05);与模型组相比,EGCG组NFκB信号通路蛋白P65、AKT信号通路蛋白AKT和MAPK信号通路蛋白P38的磷酸化水平上调(P<0.05)。综上,EGCG对免疫细胞的发育具有一定的促进以及修复作用,能够增强免疫低下小鼠的免疫功能。 To study the effect of epigallocatechin gallate(EGCG)on immune function in immunocompromised mice.An immunocompromised mouse model was constructed using cyclophosphamide,and the effects of EGCG on the immune organ index and splenic histopathological changes in immunocompromised mice were determined,and the expression levels of the immune cell nuclear transcription factors PAX5 and GATA3 as well as the expression levelsof the key molecules of the NFκB,AKT,and MAPK signaling pathways were detected.The results showed that the spleen index(1.95 g/kg)and thymus index(0.73 g/kg)of EGCG-intervened mice increased significantly(P<0.05)compared with those of the model group(1.43,0.47 g/kg),and EGCG improved the spleen mass/index of immunocompromised mice;the sectioning results showed that EGCG was able to alleviate the immune-compromised mice’s spleen injury;partially restored the expression of PAX5 in B cells and GATA3 mRNA in T cells(P<0.05);and up-regulated the phosphorylation levels of NFκB signaling pathway protein P65,AKT signaling pathway protein AKT,and MAPK signaling pathway protein P38 in the EGCG group compared with the model group(P<0.05).In conclusion,EGCG had a certain promotion as well as repairing effect on the development of immune cells,and could enhance the immune function of immunocompromised mice.
