三种真空采血管采集标本对检测总蛋白、白蛋白结果的影响

摘要 目的研究三种真空采血管对不同方法下检测总蛋白和白蛋白结果的影响。方法随机抽取80例健康体检者静脉血液样本,分别注入普通无添加剂玻璃试管(A管)、含促凝剂+分离胶试管(B管)、肝素抗凝试管(C管)中,同时用双缩脲法检测总蛋白,用溴甲酚绿法/免疫比浊法检测白蛋白。对三管结果进行统计学分析,比较其差异,从而衡量不同采血管内添加剂对双缩脲法检测总蛋白,溴甲酚绿法/免疫比浊法检测白蛋白的干扰程度。结果不同真空采血管比对实验证实,普通无添加剂玻璃试管(A管)、含促凝剂试管(B管)的双缩脲法检测总蛋白结果均低于肝素抗凝试管(C管);含促凝剂试管(B管)、肝素抗凝试管(C管)的溴甲酚绿法检测白蛋白结果均低于普通无添加剂玻璃试管(A管),差异有统计学意义(P<0.05);免疫比浊法检测白蛋白的三管间结果比较,差异无统计学意义(P>0.05)。结论总蛋白、白蛋白检测受采血管内添加剂等干扰因素影响较多。对总蛋白、白蛋白异常水平的样本建议采用更换采血管或者干扰因素少的方法如免疫比浊法进行测定,以获得可靠的结果。 Objective To study the effects of three kinds of vacuum blood collection tubes on the results of total protein and albumin detection by different methods.Methods Venous blood samples of 80 healthy subjects were randomly selected and injected into ordinary additive-free glass test tube(A tube),test tube containing coagulant+separation gel(B tube)and heparin anticoagulant test tube(C tube).At the same time,total protein was detected by biuret method,and albumin was detected by bromocresol green method/immunoturbidimetric method.The results of the three tubes were statistically analyzed to compare the differences,so as to measure the interference degree of different intravascular additives on the detection of total protein by biuret method and albumin by bromocresol green method/immunoturbidimetric method.Results The comparison experiments of different vacuum blood collection tubes confirmed that the results of total protein detection by biuret method in ordinary additive-free glass test tube(A tube)and coagulant test tube(B tube)were lower than those in heparin anticoagulant test tube(C tube);the results of albumin detection by bromocresol green method in coagulant test tube(B tube)and heparin anticoagulant test tube(C tube)were lower than those of ordinary additive-free glass test tube(A tube),and the difference was statistically significant(P<0.05).There was no significant difference in the results of albumin detected by immunoturbidimetry among the three tubes(P>0.05).Conclusion The detection of total protein and albumin is more affected by interference factors such as intravascular additives.For the samples with abnormal levels of total protein and albumin,it is recommended to use the method of replacing the blood collection tube or less interference factors such as immunoturbidimetry to obtain reliable results.
