猪丹毒丝菌的分离鉴定和耐药性分析

摘要 新疆维吾尔自治区阿克苏地区某规模化猪场猪只发生急性死亡,临床症状为关节炎和败血症等。为了解引起猪发病的致病菌及其耐药性,本试验通过实验室常规纯培养方法对采集自该猪场的病猪脏器进行细菌的分离纯化;通过革兰染色镜检和电子显微镜观察进行形态学鉴定,生化试验进行生化鉴定,并进行16S rDNA基因PCR扩增和序列分析;通过药敏试验检测分离菌株的耐药性。结果显示,分离菌株为革兰阳性菌,形态为长直、微弯曲状;生化鉴定结果显示,触酶反应、尿素、氧化酶和吲哚呈阴性,乳糖、果糖、硫化氢和葡萄糖呈阳性;结合16S rDNA基因序列比对确定分离菌株为猪丹毒丝菌。药敏试验结果显示,分离菌株对β-内酰胺类、硝基呋喃类、磺胺类和2种氨基糖苷类药物敏感。本试验结果为该规模化猪场治疗和防控猪丹毒以及进一步研究提供了科学依据。 An acute mortality event occurred in a large-scale pig farm in the Aksu region of Xinjiang Uygur Autonomous Region,with clinical symptoms including arthritis and septicemia.To identify the pathogenic pathogen and its drug resistance,routine laboratory pure culture methods were employed to isolate and purify bacteria from the affected pigs'organs collected from the farm.Morphological identification was performed through Gram staining microscopy and electron microscopy,while biochemical identification was carried out using biochemical tests.Additionally,16S rDNA gene PCR amplification and sequence analysis were conducted.Drug susceptibility of the isolated strains was tested using drug susceptibility tests.The results indicated that the isolated strains were Gram-positive,with a morphology described as long and slightly curved.Biochemical test results showed negative reactions for catalase,urea,oxidase,and indole,and positive reactions for lactose,fructose,hydrogen sulfide,and glucose.Combining these findings with 16S rDNA gene sequence alignment confirmed the isolated strains as Erysipelothrix rhusiopathiae.Drug susceptibility test revealed that the isolates were sensitive toβ-lactams,nitrofurans,sulfonamides and two aminoglycosides.These findings provide a scientific basis for the treatment and control of swine erysipelas in this large-scale pig farm and lay the groundwork for further research.
