卟啉基共价有机框架纳米粒的合成及光动力治疗研究

摘要 目的 制备卟啉基共价有机框架(COFs)纳米载体;探究纳米载体光动力抗肿瘤性质。方法 采用溶剂热法合成COFs,通过调控反应体系中催化剂用量、反应温度和反应时间,优化合成条件,进而调控COFs尺寸;对优化后的COFs纳米粒子进行表征,确定其结构。对所合成的纳米颗粒进行细胞相容性和光动力性能等性质研究。结果 所制备纳米粒子对细胞具有良好的生物相容性,且在655 nm的激光照射下,对癌细胞杀伤作用明显。在催化剂0.25 mL、反应温度为120℃、时间为48 h的条件下,获得的COFs形貌更均一、粒径最小且具有优秀的细胞相容性和光动力抗肿瘤性质。结论 通过调控制备条件成功获得COFs纳米粒子,通过MTT实验与光动力性能实验发现该纳米粒子具有良好的抗肿瘤性质。 Objective To synthesize porphyrin based covalent organic frameworks nanocarriers and explore their photodynamic therapy performance.Methods COFs were synthesized by solvothermal method.By adjusting the amount of catalyst,reaction temperature and reaction time in the reaction system,the synthesis conditions were optimized and the size of COFs was adjusted.The optimized COFs nanoparticles were characterized and the structures were determined.The cytocompatibility and photodynamic therapy properties of the synthesized nanoparticles were studied.Results All the nanoparticles exhibited good cell biocompatibility and killing effect for cancer cells under irradiation of 655 nm laser.Under the synthesis condition of 0.25 ml catalyst,120℃reaction temperature and 48 h reaction duration,the nanoparticles indicated more uniform,smallest particle size,excellent cytocompatibility and photodynamic therapy properties.Conclusion COFs nanoparticles were obtained by adjusting the preparation conditions successfully.MTT and photodynamic experiments showed that the nanoparticles possess good antitumor properties.
