黄芪源性毛蕊异黄酮与游泳训练协同作用改善AD大鼠的认知功能

The Synergistic Effect of Astragalus-Derived Calycosin and SwimmingTraining Improves Cognitive Function in AD Rats

摘要 黄芪(Astragalus membranaceus)是一种传统中药材,毛蕊异黄酮(calycosin,CAL)是黄芪中含量丰富的异黄酮类化合物,具有改善记忆障碍的作用。以往研究证实游泳与药物疗法对于AD的治疗更有效,本研究旨在探讨黄芪源性毛蕊异黄酮与游泳训练的协同作用,以及其通过激活海马BDNF/TrkB/CREB信号通路改善AD大鼠认知功能的可能性。通过实验研究,我们发现毛蕊异黄酮联合游泳训练能够显著提高AD大鼠海马区BDNF、TrkB和p-CREB蛋白的表达水平,减轻海马损伤和细胞凋亡,降低Aβ1-42和BACE1的表达,减轻氧化应激和炎症状态。这些结果表明,毛蕊异黄酮与游泳训练通过激活BDNF/TrkB/CREB信号通路,协同改善AD大鼠的认知功能,为AD的非药物治疗提供了新的思路。本研究不仅揭示了黄芪源性毛蕊异黄酮与游泳训练的潜在治疗价值,也为未来相关疾病的治疗研究提供了重要的科学依据。 Astragalus membranaceus is a traditional Chinese medicinal herb,and calycosin(CAL)is an isoflavone compound rich in Astragalus membranaceus known for its memory improvement effects.Previous research has confirmed that swimming combined with pharmacotherapy is more effective in treating Alzheimer's disease(AD).This study aims to explore the synergistic effects of calycosin derived from Astragalus membranaceus and swimming training,and its potential in improving cognitive functions in AD rats through the activation of the hippocampal BDNF/TrkB/CREB signaling pathway.Experimental studies have shown that calycosin combined with swimming training significantly enhances the expression levels of BDNF,TrkB,and p-CREB proteins in the hippocampus of AD rats,alleviates hippocampal damage and cell apoptosis,reduces the expression of Aβ1-42 and BACE1,and mitigates oxidative stress and inflammation.These results indicate that calycosin and swimming training,by activating the BDNF/TrkB/CREB signaling pathway,synergistically improve cognitive functions in AD rats,providing a new approach for non-pharmacological treatment of AD.This study not only reveals the potential therapeutic value of calycosin derived from Astragalus membranaceus and swimming training but also provides a significant scientific basis for future research on the treatment of related diseases.
