摘要
本研究旨在探讨柴胡(Bupleurum chinensis DC.)活性物质柴胡皂苷A(Saikosaponin A,SSA)联合有氧运动对肥胖大鼠的治疗作用及其机制。通过构建肥胖大鼠模型,研究柴胡皂苷A与有氧运动的单独及联合应用对肥胖大鼠体重、血脂水平、肝脏脂肪积累及脂肪生成相关基因表达的影响。结果显示,与单独治疗相比,柴胡皂苷A联合有氧运动显著降低了大鼠的体重和肝脏脂肪积累,改善了血脂参数,并通过上调抗氧化酶活性和下调促炎细胞因子水平表达发挥抗氧化和抗炎作用。进一步机制研究表明,此联合治疗可激活Nrf2通路,抑制NF-κB通路,有效调节脂肪生成相关基因的表达。本研究证实了柴胡皂苷A联合有氧运动在肥胖治疗中的协同效应,为肥胖的综合治疗提供了新的策略和见解。
This study aims to explore the therapeutic effects and mechanisms of the active substance Saikosaponin A(SSA)from Bupleurum chinensis,in combination with aerobic exercise on obese rats.An obesity rat model was established to examine the effects of both individual and combined applications of Saikosaponin A and aerobic exercise on the weight,blood lipid levels,liver fat accumulation,and the expression of genes related to fat generation in obese rats.Results indicated that compared to individual treatments,the combination of Saikosaponin A and aerobic exercise significantly reduced the weight and liver fat accumulation in rats,improved lipid parameters,and exerted antioxidant and anti-inflammatory effects by upregulating the activity of antioxidant enzymes and downregulating pro-inflammatory cytokine levels.Further mechanistic studies revealed that this combined treatment activates the Nrf2 pathway and inhibits the NF-κB pathway,effectively regulating the expression of genes associated with fat generation.This study confirmed the synergistic effects of Saikosaponin A and aerobic exercise in the treatment of obesity,providing new strategies and insights for comprehensive obesity management.
