摘要
目的 探讨烧烫伤外用中药专利复方的用药规律和可能作用机制。方法从中国专利数据库中筛选治疗烧烫伤的外用中药专利复方,利用古今医案云平台对纳入研究的复方进行频数、聚类和关联分析;通过网络药理学预测外用中药复方治疗烧烫伤的活性成分及治疗靶点,并对潜在靶点进行通路富集分析,然后通过分子对接验证关键成分与核心靶点的结合力。结果共发现治疗烧烫伤的外用中药专利复方274个,涉及中药621味;最常用的药物是冰片、地榆、大黄等,主要功效为清热解毒,药多苦寒,归肝经;从使用频次前5的中药中筛出核心活性成分有槲皮素(Quercetin)、山奈酚(Kaempferol)、β-谷甾醇(β-sitosterol)等,核心靶点有肿瘤坏死因子(TNF)、白细胞介素-6 (Interleukin-6,IL-6)、丝氨酸/苏氨酸蛋白激酶1 (AKT1)、白细胞介素-1β(1L-1β)、血管内皮生长因子A (VEGFA)等,这些靶点可通过干预白细胞介素17(IL-17)、磷脂酰肌醇-3激酶-蛋白激酶B (PI3K-Akt)、丝裂原活化蛋白激酶(MAPK)、TNF、低氧诱导因子-1 (HIF-1)等信号通路起效;分子对接显示核心成分与核心靶点均有较好的结合力。结论外用中药治疗烧烫伤可通过提取中药活性成分,靶向调控炎性因子及生长因子的表达,多途径促进创面愈合,亦为中药外用制剂的开发提供思路。
Objective This study aimed to investigate the therapeutic principles and mechanisms of action of patented traditional Chinese medicine(TCM)formulations for topical application in the management of burns and scalds.Methods Patented formulations of topical Chinese medicine for burns and scalds were retrieved from the Chinese patent database.These formulations underwent frequency,clustering,and correlation analyses using the Ancient and Modern Medical Records Cloud Platform.Network pharmacology was employed to predict the active constituents and therapeutic targets of topical TCM formulations for burns and scalds.Pathway enrichment analysis was performed to identify potential targets,followed by molecular docking to validate the binding affinity between key constituents and core targets.Results A total of 274 patented compound formulas,involving 621 types of Chinese medicinal materials,were found for external treatment of burns and scalds.The most commonly used drugs are borneol,lithospermum,and rhubarb,with the main effects of clearing heat and detoxification.The medicines are mostly bitter and cold,belonging to the liver meridian.Core active ingredients such as quercetin,kaempferol,andβ-sitosterol were identified from the top 5 Chinese medicines by frequency of use,with core targets including tumor necrosis factor(TNF),interleukin-6(IL-6),serine/threonine kinase 1(AKTI),interleukin-1β(IL-1β),vascular endothelial growth factor A(VEGFA),and others.These targets can be effective through intervention in signaling pathways such as interleukin-17(IL-17),phosphoinositide 3-kinase-protein kinase B(PI3K-Akt),mitogen-activated protein kinase(MAPK),TNF,hypoxia-inducible factor-1(HIF-1),etc.Molecular docking shows that the core components have good binding affinity with the core targets.Conclusion External application of TCM for treating burns and scalds can promote wound healing through the extraction of active ingredients,targeting the regulation of inflammatory factors and growth factor expression,thereby providing insights for the development of topical Chinese medicine formulations.
