鱼腥草素钠对高血压大鼠模型血压的影响及其作用机制

摘要 目的探讨鱼腥草素钠对高血压大鼠模型血压的影响及其作用机制。方法选择8周龄的雄性Sprague-Dawley大鼠,随机分为对照组(n=11)、高血压组(n=13)和鱼腥草素钠组(n=13)。对照组采用普通饲料喂养,高血压组和鱼腥草素钠组均采用高脂饲料喂养,建立高血压模型。测量大鼠基线血压,每周监测1次大鼠体重,从第5周起监测大鼠尾动脉血压,第9周起鱼腥草素钠组大鼠采用鱼腥草素钠灌胃,对照组及高血压组大鼠采用等量生理盐水灌胃。第13周后检测3组大鼠血浆炎症因子[白介素1β(IL-1β)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)]水平及血脂、血糖、胰岛素浓度。结果对照组收缩压和鱼腥草素钠组相比,差异无统计学意义(P>0.05),但均低于高血压组(P<0.05);鱼腥草素钠组和高血压组血浆总胆固醇水平均高于对照组,差异有统计学意义(P<0.05);鱼腥草素钠组IL-1β、IL-6和TNF-α水平明显低于高血压组,差异有统计学意义(P<0.05),而与对照组比较,差异无统计学意义(P>0.05)。结论鱼腥草素钠干预后,可明显缓解炎症,降低高脂喂食大鼠导致的高血压。 Objective To investigate the effect of sodium houttuyfonate(SH)on the blood pressure in hypertensive model rats and its potential mechanism.Methods A total of 37 male Sprague-Dawley rats aged 8 weeks were randomly divided into control group(n=11),hypertension group(n=13)and SH group(n=13).The control group was fed with ordinary diet,while the hypertension group and SH group were fed with high-fat diet to establish hypertension model.The baseline blood pressure was measured,the weight was monitored weekly,and the blood pressure of the tail artery of the rats was monitored from the 5th week.From the 9th week,the rats in the SH group were given SH gavage,while rats in the control group and the hypertension group were given the same amount of normal saline gavage.The levels of plasma inflammatory factors interleukin 1β(IL-1β),interleukin 6(IL-6),and tumor necrosis factorα(TNF-α),blood lipid,blood glucose,and insulin in the 3 groups were detected at the end of week 13.Results There was no significant difference in systolic blood pressure between the control group and the SH group(P>0.05).However,the systolic blood pressure in the control group and the SH group was lower than that in the hypertension group(P<0.05).The level of plasma total cholesterol in the SH group and the hypertension group was higher than that in the control group(P<0.05).The levels of IL-1β,IL-6 and TNF-αin the SH group were lower than those in the hypertension group(P<0.05),but no significant difference was found in the above indicators between the SH group and the control group(P>0.05).Conclusion SH can significantly alleviate inflammation and reduce hypertension induced by high-fat diet in rats.
