江山黄精化学成分UPLC-Q/TOF-MSE分析及其抗氧化和抗炎活性研究

UPLC-Q/TOF-MS E Analysis of the Chemical Composition of Jiangshan Polygonatum Sibiricum and Its Antioxidant and Anti inflammatory Activities

摘要 [目的]探讨江山黄精醇提物(Jiangshan Polygonatum sibiricum alcoholic extract,PSAE)的化学成分、抗氧化和抗炎活性。[方法]]使用超高效液相色谱-四极杆-飞行时间质谱(ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry,UPLC-Q/TOF-MSE)分析PSAE化学成分,测定其抗氧化活性,体外实验测定其对脂多糖(lipopolysaccharide,LPS)诱导的THP-1细胞的影响,并使用分子对接进行验证。[结果]PSAE主要含有17种化学成分,包括黄酮类化合物5种、皂苷类化合物5种、木脂素类化合物3种、酚酸类化合物1种、甾体类化合物1种,其他类化合物2种。体外实验表明PSAE具有良好的体外抗氧化活性,能增加LPS诱导的THP-1细胞活力,显著降低细胞上清液中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)和白细胞介素-1β(interleukin-1β,IL-1β)的含量及NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor thermal protein domain associated protein 3,NLRP3)、凋亡相关斑点样蛋白(apoptosis-associated speck-like protein containing CARD,ASC)和胱天蛋白酶-1(cysteinyl aspartate specific proteinase-1,Caspase-1)的表达。分子对接结果显示,17种化学成分中的16种均与核心靶点NLRP3、ASC和Caspase-1有良好的结合活性。[结论]PSAE具有较好的抗氧化活性和显著的抗炎活性,其发挥作用的主要活性成分可能为黄酮及皂苷类化合物。 [Objective Objective]To investigate the chemical compositions,antioxidant and anti-inflammatory activities of Jiangshan Polygonatum sibiricum alcoholic extract(PSAE)in vitro.[Methods Methods]The chemical compositions of PSAE were identified by ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q/TOF-MS E),its antioxidant activity was studied,and in vitro experiments were conducted to determine its effect on lipopolysaccharide(LPS)-induced THP-1 cells.Molecular docking was further used to validate the results.[Results Results]PSAE contained 17 chemical compositions,including 5 flavonoids,5 saponins,3 lignans,1 phenolic acid,1 steroid and 2 other compositions.In vitro experiments showed that PSAE possessed great antioxidant activity.Besides,PSAE could increase the viability of LPS-induced THP-1 cells,significantly decrease the levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in cell supernatants and the expression of NOD-like receptor thermal protein domain associated protein 3(NLRP 3),apoptosis associated speck-like protein containing CARD(ASC)and cysteinyl aspartate specific proteinase-1(Caspase-1).Molecular docking results showed that 16 of the 17 chemical compositions detected in PSAE had good docking activities with the core targets of NLRP 3,ASC and Caspase-1.[Conclusion Conclusion]PSAE has great antioxidant and anti-inflammatory activities,and the main active compositions may be flavonoids and saponins.
